Journal ArticleDOI
Tumorigenicity and genetic profiling of circulating tumor cells in small-cell lung cancer
Cassandra L Hodgkinson,Christopher J. Morrow,Yaoyong Li,Robert Metcalf,Dominic G. Rothwell,Francesca Trapani,Radoslaw Polanski,Deborah J. Burt,Kathryn Simpson,Karen Morris,Stuart D Pepper,Daisuke Nonaka,Alastair Greystoke,Paul P. Kelly,Becky Bola,Matthew G Krebs,Jenny Antonello,Mahmood Ayub,Suzanne Faulkner,Lynsey Priest,Louise Carter,Catriona Tate,Crispin J. Miller,Fiona H Blackhall,Ged Brady,Caroline Dive +25 more
TL;DR: It is shown that CTCs from patients with either chemosensitive or chemorefractory SCLC are tumorigenic in immune-compromised mice, and the resultant CTC-derived explants (CDXs) mirror the donor patient's response to platinum and etoposide chemotherapy.
Journal ArticleDOI
The Histone Demethylase KDM1A Sustains the Oncogenic Potential of MLL-AF9 Leukemia Stem Cells
William J. Harris,Xu Huang,James T. Lynch,Gary J. Spencer,James R. Hitchin,Yaoyong Li,Filippo Ciceri,Julian G. Blaser,Brigit F. Greystoke,Allan M. Jordan,Crispin J. Miller,Donald J. Ogilvie,Tim C. P. Somervaille +12 more
TL;DR: The data establish KDM1A as a key effector of the differentiation block in MLL leukemia, which may be selectively targeted to therapeutic effect, and drugs active in the nanomolar range phenocopied Kdm1a knockdown in both murine and primary human AML cells exhibiting MLL translocations are established.
Journal ArticleDOI
Molecular analysis of circulating tumor cells identifies distinct copy-number profiles in patients with chemosensitive and chemorefractory small-cell lung cancer
Louise Carter,Dominic G. Rothwell,Barbara Mesquita,Christopher Smowton,Hui Sun Leong,Fabiola Fernandez-Gutierrez,Yaoyong Li,Deborah J. Burt,Jenny Antonello,Christopher J. Morrow,Cassandra L Hodgkinson,Karen Morris,Lynsey Priest,Mathew Carter,Crispin J. Miller,Andrew Hughes,Fiona H Blackhall,Caroline Dive,Ged Brady +18 more
TL;DR: Significant difference was observed in progression-free survival (PFS) between patients designated as chemorefractory or chemosensitive by using the baseline CNA classifier, which suggests that the genetic basis for initial chemoresistance differs from that underlying acquired Chemoresistance.
Journal ArticleDOI
GFI1 proteins orchestrate the emergence of haematopoietic stem cells through recruitment of LSD1
Roshana Thambyrajah,Milena Mazan,Milena Mazan,Rahima Patel,Victoria Moignard,Monika Stefanska,Elli Marinopoulou,Yaoyong Li,Christophe Lancrin,Thomas Clapes,Tarik Möröy,Catherine Robin,Crispin J. Miller,Shaun M. Cowley,Berthold Göttgens,Valerie Kouskoff,Georges Lacaud +16 more
TL;DR: It is established that in the absence of GFI1 proteins, HSCs and haematopoietic progenitor cells are not produced in the AGM, revealing the critical requirement for GFI 1 proteins in intra-embryonic EHT.
Proceedings Article
The Perceptron Algorithm with Uneven Margins
TL;DR: The interest of these algorithms from a theoretical point of view is discussed, a generalisation of Novikoff’s theorem for uneven margins is provided, and both algorithms yield equal or better performances than support vector machines, while reducing training time and sparsity.
