Introduction : The gamma-aminobutyric acid type A (GAB ) receptor is an important pharmacological target of alcohol. The phamacological characteristics of the receptor are largely determined by its subunit composition. Compared with all other subtypes, the 6- containing receptors are more sensitive to GABA and less sensitive to benzodiazepines.The purpose of this study was to address a role for GAB 6 receptor subunit gene in the development of alcohol dependence. The differential manifestation of alcohol withdrawal symptoms related to GAB 6 polymorphism in patients treating with benzodiazepines was also examined. Methods: Eighty-seven inpatients with alcohol dependence, and sixty healthy controls were evaluated using CIWA-Ar scale. Each patient was genotyped for GAB 6 subunit. Association between GAB 6 polymorphism and severity of withdrawal symptom were determined. Results : No significant difference was found in GAB 6 receptor genetic type and allelic distribution between the alcohol dependent and control subject. Tremor was more severe in CC than TT type. TT type had higher degree of anxiety, agitation and headache than CC type. The GAB 6 C allele increased the average score of tremor significantly, and T allele increased that of agitation. Conclusion : The results suggested that GAB 6 genetic polymorphism was not associated with alcohol dependence and with severity of alcohol withdrawal symptoms. But in benzodiazepine treated patients, GAB 6 polymorphism and allelic type show the difference in severity of each withdrawal symptom. These differences of severity are partly responsible for the unique pharmacological properties associated with the GAB 6 subunit.

Treatment of Intertrochanteric Fracture of Femur: A Randomized Prospective Comparative Analysis of the Internal Fixation of Gamma Nail and Compression Hip Screw

The abnormal deposition of amyloid b peptide (AbÞ is a hallmark of Alzheimer’s disease (AD). Phospholipase C-d1 (PLC-d1) is also known to abnormally accumulate in the brains of AD patients, but no report has addressed the relationship between these two events. This study investigated the effect of Ab42 on the PLC-d1 expression in human neuroblastoma cell lines. The PLC-d1 mRNA level was increased by treatment with Ab42 in a RT-PCR analysis. In the reporter assay, Ab42 was found to activate the PLC-d1 promoter activity in a dose-dependent manner. A novel NF-jB binding site in the PLC-d1 promoter appeared to be responsible for the Ab42 activity. First, the dominant negative forms of the NF-jB activating molecules, dominant negative TGF-b activated kinase 1 (dnTAK1) and dnNIK (dominant negative NF-jB-inducing kinase), abolished the Ab42 activity in the reporter assay. Second, the Ab42 augmented a factor binding on the NF-jB site in the electrophoretic mobility shift assay (EMSA), which was abolished by a molar excess of the unlabeled consensus NF-jB oligonucleotide. These results suggest that the PLC-d1 promoter is under the control of NF-jB, which mediates the expression of PLC-d due to the Ab42 treatment. 2003 Elsevier Inc. All rights reserved.
