A continuous fluorescence-based assay for the human high-molecular-weight cytosolic phospholipase A2.

TL;DR: A sensitive method for continuously monitoring the activity of the human cytosolic phospholipase A2 (cPLA2) is described and it is demonstrated that when 7-hydroxycoumarinyl gamma-linolenate was dispersed in aospholipid matrix it was hydrolyzed by cPLA2 at a rate comparable to that of an arachidonyl-containing phospholIPid substrate and with an identical reaction progress curve.

TL;DR: A series of naphthalenebis(difluoromethylenephosphonates) were prepared and compared to their monosubstituted counterparts as inhibitors of the protein phosphatases, PTP1B, CD45 and PP2A and some of these are among the most potent, nonpeptidyl inhibitors of protein tyrosine phosphatased substances reported to date.

SAR and optimization of thiazole analogs as potent stearoyl-CoA desaturase inhibitors

TL;DR: Compound 3j, a potent SCD inhibitor was identified from the optimization of a lead thiazole compound MF-152 with over 100-fold improvement in potency and gave a robust 24% prevention of body weight gain in mice fed on a high fat diet accompanied with an improved metabolic profile on insulin and glucose levels.

Human cytosolic phospholipase A2 expressed in insect cells is extensively phosphorylated on Ser-505

TL;DR: Sf9 cells have a mechanism for phosphorylating cPLA2 similar to that found in mammalian cells which probably proceeds through a MAP kinase, thus, insect cell overexpressed c PLA2 is a very good source for the Ser-505 phosphorylated enzyme.
