Abstract: Two cathepsins were detected in Mujil auratus muscle extracts. They were classified as a thiol- and aspartyl-proteinase (cathepsins B and D, respectively) on the basis of their catalytic behaviour in presence of specific inhibitors. 1. 2. Following extraction in 1% KCl, the proteinases were purified by autolysis, acetone fractionation, affinity chromatography, and gel permeation chromatography. The haemoglobin-agarose column chromatography allowed us to separate the two activities. 2. 3. Sephadex G-75 column chromatography resulted in apparent molecular weights of 25,000 (cathepsin B) and 35,000 (cathepsin D). 3. 4. The molecular size, together with pH-activity profiles and kinetic parameters are similar to those reported for mammalian cathepsins B and D. 4. 5. This was not the case with the temperature-activity profiles, the optimum temperature as well as the heat stability being higher for fish cathepsins than for those obtained from other sources. 5. 6. Cathepsin B was characterized by its ability to inactivate aldolase. 6. 7. Fluorescence quenching experiments showed that tryptophyl residues of cathepsin B were less occluded and located in a more electronegative microenvironment that those pertaining to cathepsin D.

Abstract: Acid catheptic activity was measured in crude extracts of muscle, liver, heart, spleen and gonads from the fishes Mujil auratus, Sparus aurata and Lightonatus mormyrus. The spleen was the organ which showed the highest activity. A comparative study of the seven most commonly used extraction methods was made. Some were modified to account for the characteristics of the fish organs and the activity extracted from them. The Siebert method resulted as the best extraction method only if 1 mM EDTA was present in the medium. The activity from Mujil auratus muscle was strongly inhibited by iodoacetate, N-ethylmaleimide, p-hydroxy mercuribenzoate, and diazo-acetyl-DL-norleucine methyl ester. The results indicated the presence of a carboxyl-proteinase and a thiol-proteinase. According to inhibition studies, the levels of proteinase and amidase activities shown by different organs of Mujil auratus were re-examined. The spleen extract showed the maximum activity for both cathepsins, but muscle extract accounted for more than 95% of total catheptic activity.

Abstract: The evolution of microporosity of activated carbons prepared from carbonized olive stones as a function of burn-off in carbon dioxide (activating agent) has been studied at high temperature using a gas chromatographic technique. The specific net retention volumes have been used to determine the possible molecular sieve properties of the carbons towards several hydra-carbons. This molecular sieving is thought to be due to the existence of slit-shaped pores in the carbons with well-defined dimensions. The retention data have also been used to calculate the heats of adsorption and their dependence on the porosity of the carbons.

Abstract: Fracturing is the most conspicuous type of tectonics developed in the SW of Sierra Nevada after the alpine building of the Betic Cordilleras. Fractures have been inspected on satellite images as well as on ordinary photograms and surveyed through field studies, including microtectonic measurements in several favorable sectors, There are three main sets of fractures. N 70-100 E, N 120-150E and N 10-30E. Although they were likely strike-slip faults in origin, they show numerous movements, both horizontal and vertical, ranging possibly up to the present time more, effectively controlling the sedimentation during the Neogene and Quaternary and being largely responsible for the present relief. The horizontal movements took place under a compressional stress trending essentially N 120 E and approximately N-S, acording to the results of microtectonic studies.

Abstract: The effect of strong visible light on spores of Bacillus subtilis has been studied. With the illumination conditions used, spores were killed exponentially and viability decreased to less than 13% survivors after 24 h. This decrease did not take place when the cells were kept under strictly anaerobic conditions. The killing is, therefore, attributable to a photodynamic effect. The presence in the medium of exogenous photosensitizers did not increase the effect of light and this is attributable to the impermeability of spores to these agents. Once germinated, the spores were killed at a much faster rate, even when outgrowth was stopped. The highest sensitivity was reached 90 min after onset of outgrowth.

Abstract: Subcellular distribution studies of calmodulin in cat sympathetic ganglia demonstrated that about 90% of the protein remained in the 27,000 g supernatant, suggesting that it is a cytosolic protein. Only 4.5% was recovered in the microsomal fraction pellet. The inferior mesenteric ganglia contained 93.3 +/- 3 ng calmodulin per ganglion, and segments of unligated cat hypogastric nerves had 6.53 +/- 0.32 ng per 5 mm segment. When the nerve was ligated in the middle and left in the cat for 1-6 days, substantial amounts of calmodulin accumulated in segments of nerve immediately proximal (P1) and distal (D1) to the ligature. The amounts found in P1 amounted to 15.3, 20, 30.4 and 39.4 ng calmodulin per 5 mm segment 1, 2, 3 and 6 days after ligation, respectively. The average rate of transport was 5.5 mm per day, which corresponds to a slow component b of axonal transport (SCb). The accumulation of calmodulin in D1 was also increased with the time of ligation. After 1, 2, 3 and 6 days, the amounts of the protein found in D1 were 14.4, 17.7, 19 and 21 ng per 5 mm segment, respectively. The calculated mean rate for the retrograde transport was 3.9 mm per day. Decentralization of the inferior mesenteric ganglia did not affect the rate of accumulation of calmodulin or the basal amounts found in ganglia and nerves. Local injection inhibited the orthograde, but not the retrograde axonal transport of the protein. It is concluded that calmodulin undergoes a process of slow orthograde axonal transport probably incorporated into the axoplasmic matrix of a network of actin microfilaments. The protein is also transported in a retrograde manner.

Abstract: A purple mutant of Halobacterium halobium was isolated in a previous study. The ‘in vitro’ absorption spectra of the cells gave a broad shoulder around 570 nm. The amounts of bacteriorhodopsin were high under any growth condition (including aerobic) inhibitory for the wild-type strain. The mutant grew faster under illuminated microaerophilic conditions and showed faster proton extrusion than the wild-type strain. This evidence shows that the mutant has a constitutive bacteriorhodopsin production not influenced by the oxygen concentration in the medium. However, some stimulation by light was found.

Abstract: Hemos querido retomar, para este numero monografico, la obra del ilustrado italiano L A. Muratori, valorandola, en esta ocasion, desde la perspectiva de la labor de traduccion de una parte de sus obras que se llevo a cabo en Espana durante la segunda mitad del siglo XVIII. Lo cual, pensamos, permitira por una parte contribuir a un mayor conocimiento de las obras que pudo leer no la elite ilustrada—que por su propia formacion podia acceder con relativa facilidad a las mas novedosas producciones intelectuales que se iban editando en Europa—, sino la masa de espanoles a la que se le franqueo la posibilidad de conocer parcialmente dicha produccion por medio de las traducciones. Por otra, el presente estudio permitira concretar algo mas la tesis—ya indiscutiblemente aceptada—que contempla la participacion activa de una corriente de influencia italiana en el proceso de gestacion y desarrollo del movimiento ilustrado espanol.

Abstract: La caracteristica fundamental de la compania que el 30 de noviembre de 1759 formaron en Valencia cinco mercaderes de libros y tres impresores, deriva del tipo de asociacion que adoptaron, es decir, el constituir una sociedad por acciones. El desarrollo de este tipo de sociedades en la Espana del siglo XVIII resulta de enorme interes, puesto que contrasta con la forma mas habitual con la que los comerciantes solian unir sus capitales y esfuerzo para la consecucion de sus fines, utilizando un tipo de companias que eran conocidas como generales o «colectivas», o, con menor frecuencia, las comanditarias.

Abstract: Long-term exposure to hexane and acute or accumulative chronic ingestion of many organophosphates induce peripheral neuropathies (Spencer et al. 1980; Johnson 1982). Many cases of similar peripheral neuropathies have been reported in workers handling solvents or adhesives in leather factories in Italy, Spain, Japan and other countries (Vilanova 1981). Many of them have been attributed either to hexane or organophosphates (mainly TOCP). This laboratory has observed a possible potentiation of the hexane and organophosphate neurotoxicities. Hens were exposed to hexane at doses producing slight weakness, but not ataxia. They devolped a severe ataxia (3–4 points in a 4-points scale) after a single TOCP dose which induced only a slight ataxia in the hexane non-exposed hens (1–2 points). The possibility that the impurities in the industrial solvents could play a significant role is shown by the observation that the non-hydrocarbon impurities in many samples of commercial hexane inhibited S-ChE “in vitro” but not AChE. An inhibition without preincubation is observed and also a time progressive inhibition with kinetic properties in accord with organophosphate inhibitors. Organic elemental phosphorus has been measured. Several fractions have been isolated by preparative chromatography. In one of them dibutyl, hexyl and dioctyl phthalates, adipates and triphenylphosp-hate have been identified by GC-mass spectrometry. The presence of dibutylphthalate could explain the non-progressive inhibition. The progressive inhibition is not explained by the triphenylphosphate. Another unidentified organophosphate has to be in the hexane impurities to explain the non-progressive inhibition. The potentiation effect of organophosphates and hexane, besides the presence of organophosphates and inhibitors of sterases are facts to take into consideration in understanding a multifactor explanation of occupational toxic neuropathies.

Abstract: No es preciso que indiquemos la importancia que adquieren, en la investigacion de los fenomenos culturales, una serie de estudios: procesos de alfabetizacion e instruccion (y su incidencia en las distintas capas sociales), analisis de bibliotecas (tanto de particulares como de instituciones), sociologia de los lectores, etc. No obstante, si estos trabajos apenas se han iniciado a nivel nacional, su carencia es absoluta respecto de Murcia. El aspecto mejor conocido es precisamente la produccion editorial, y en torno a ella desarrollamos el presente articulo. Su valor como reflejo e indicador de una realidad cultural, como marco de referencia en el que situar o valorar las corrientes dominantes (y la aparicion de otras nuevas), no ofrece dudas. Quede claro, por otra parte, que se trata de una aproximacion parcial, cuyos resultados deben contrastarse con los obtenidos en otras investigaciones que mencionaremos en este articulo.

Abstract: La idea de crear una biblioteca destinada a tener una funcion publica cuenta con importantes precedentes entre nuestros humanistas del siglo XVI, pero aqui mas que de bucear en ellos, y en su fracaso, nos ocuparemos de marcar algunas lineas que contribuyeron a que un proyecto llegase a cuajar en un Real Decreto de constitucion, asi como de senalar algunos de los errores comunes que se han ido repitiendo en torno a los origenes de la Biblioteca Nacional. Las referencias a los fondos iniciales de la Biblioteca rara vez aparecen separados de los acontecimientos de la Guerra de Sucesion y se identifican con el tema de las confiscaciones a los disidentes del partido borbonico. Maldonado Macanaz senalaba como entre los libros que formaron el primer nucleo se encontraban «los dos mil volumenes de la libreria de Fray Antonio Folch de Cardona». En principio, la afirmacion no deja de ser cierta, pero en realidad la biblioteca personal del arzobispo estuvo muy poco tiempo en las estanterias de la Biblioteca Real como se podra comprobar en las paginas siguientes.
