Abstract: Objective It has become more important to protect brain function in operation and post-operation period with the developmen of surgery therapy for congenital heart surgery.Methods The primary cultured neuron cell of l8-day-pregnan embryoid Sprague-Dawley rat's hippocampus has been selected as the experimental material.The experimental concentration of magnesium sulfate will be 0 mmol/L,1 mmol/L,2 mmol/L,4 mmol/L;0 mmol/L is for control group.Selecting c-fos protein as target for primary antibody.Results After immunohistochemistry procedure,we found the mortality of neuronal cultures with magnesium sulfate in different concentration was 89.47 %,49.35 %,52.56 %,90.67 %(P0.01).Conclusions The results reiterate the neuroprotection of magnesium sulfate.The results also show the effective time of magnesium sulfate is 2 to 4 hours. After 4 to 8 hours,the neuroprotection has become faint.

Abstract: Objective To evaluate the in vitro cytokine-producing ability and the immunoprotection of the mice immunized with the TB DNA vaccine pVS3in1.Methods Plasmid pVS3in1 expressing fusion protein Mtb8.4-38 000-Ag85B of Mycobacterium tuberculosis was constructed by inserting mtb8.4,CTL epitope of 38 000,and ag85b into the vector pVAX1.One hundred female inbred C57BL/6 mice were divi-(ded) into 5 groups,and then immunized respectively with pVS3in1,pIL2S,pVAX1,PBS,and BCG for 3 times with 2 weeks intervals except BCG group(one injection only).Sera and supernatants of 10 mice in each group were tested for determining hIL-2,mIL-2,mIFN-γ,mIL-6,and mIL-10 by sandwich ELISA.The other 10 mice in each group were challenged with 10~6 CFUs M.tuberculosis H37Rv and sacrificed for culturing H37Rv from the spleens,lungs,and livers.Results The average concentrations of mIL-2 and mIFN-γ in the supernatants of the mic from pVS3in1 immunized mice were(379.6±58.2)pg/mL and(529.7±63.7)pg/mL,respectively,which were significantly higher than those from the pVAX1,pIL2S,and PBS injected controls(P0.001).No significant difference found in the mIL-6 and mIL-10 concentrations of the mice from all 5 groups.The average loads of M.tuberculosis in the spleens,livers and lungs in the pVS3in1 group were((17 443.6±3 202.5)CFU/g),(19 047.2±3 395.5)CFU/g and(14 822.2±2 882.2) CFU/g,respectively.The average bacterial loads of the 3 organs in the pVS3in1 immunized group were significantly lower than those of their counterparts in pVAX1,pIL2S and PBS groups,but bacterial load in the spleens was significantly higher than that of their counterparts in the BCG immunized controls.Conclusions TB DNA vaccine pVS3in1 can induce Th1 immune response,which is necessary for prevention against TB,and its ability to enhance immunoprotection against H37Rv challenge in the immunized mice is equivalent to BCG vaccinization.

Abstract: We developed a Web-based system to interactively display image-based electronic patient records (EPR) for intranet and Internet collaborative medical applications. The system consists of four major components: EPR DICOM gateway (EPR-GW), Image-based EPR repository server (EPR-Server), Web Server and EPR DICOM viewer (EPR-Viewer). We have successfully used this system two times for the teleconsultation on Severe acute respiratory syndrome (SARS) in Shanghai Xinhua Hospital and Shanghai Infection Hospital. During the consultation, both the physicians in infection control area and the experts outside the control area could interactively study, manipulate and navigate the EPR of the SARS patients to make more precise diagnosis on images with this system assisting. This presentation gave a new approach to create and manage image-based EPR from actual patient records, and also presented a way to use Web technology and DICOM standard to build an open architecture for collaborative medical applications.

Abstract: OBJECTIVE To investigate the possibility of using amniotic fluid cells as seed cells for tissue engineering. METHODS Amniotic fluid was obtained by ultrasound-guided amniocentesis performed on pregnant women with a gestational age ranging from 16(th) approximately 23(rd) weeks. The cells isolated from the amniotic fluid were cultured in F10 culture fluid with 10% FBS. Immunocytochemistry was used to examine the standard intermediate filaments. After 3 passages of subculture, the cells were harvested and seeded onto PGA polymer scaffold. The cellular morphology, structure and adhesion with scaffold were evaluated by contrast microscope and scanning electronic microscope. RESULTS The amniocytes expanded rapidly in culture media. Immunocytochemistry revealed positive signals for vimentin, smooth muscle action (SMA), and pan cytokeratin, and negative signals for desmin. Amniocytes-polymer complex analysis showed confluent cells firmly attached to the scaffold, with no evidence of cell death. CONCLUSION The expansion potential of amniotic fluid cells is active. They express the characteristics of mesenchymal cells. The cells on PGA polymer scaffold can grow rapidly and maintain its morphological property. So the amniotic fluid may be a practical cell source for tissue engineering.

Abstract: Objective:To explore the clinical efficacy of ABCD2 score in predicting transformation of transient ischemic attack(TIA) into cerebral infarction(CI) within 7 days. Methods: According to occurrence of CI 7 days after TIA, 115 TIA patients were divided into CI group(n=33) and non-CI group(n=82). Risk factors of cerebrovascular diseases and ABCD2 scores were documented. Single factor and multivariate analysis have been used to explore the differences in risk factors and ABCD2 score contents between the two groups. Receiver operating characteristic curve(ROC) analysis was used to explore the efficacy of ABCD2 score as well. Results: The age and ABCD2 score in the CI group were significantly higher than those in the non-CI group(P0.05 or P0.01). On single factor analysis showed that, in the CI group, the proportion of patients older than 60 years, systolic blood pressure higher than 140 mmHg and/or diastolic pressure higher than 90 mmHg, unilateral limb force weak, duration of symptoms longer than 60 min were significantly higher than that in the non-CI group(P0.05). In addition, in the CI group, the proportion of patients whose symptoms persisted for 10~59 min was much less than that in the non-CI group(P0.05). Multivariate analysis showed the same results. ROC analysis showed that ABCD2 score of 4.5 was promised point in predicting TIA evolving into CI within short-term. The area under curve was 0.73 with sensitivity of 57.6% and specificity of 78.0%, Conclusion: ABCD2 score was a simple and effective tool to predict whether TIA will evolve into CI in short-term. ABCD2 score above 5 might have an important predictive value.
