Abstract: [Objective]To investigate the single nucleotide polymorphism-1131 T/C of the apolipoprotein A5(APOA5)gene possible association with coronary artery disease(CAD)and its distribution in Chinese Han population.[Methods]The single nucleotide polymorphisms(SNP)at positions1131 T/C in APOA5 gene were determined by polymerase chain reaction-restriction fragment length polymorphisms(PCR-RFLP)assay in persons of Han nationality in Nanjing district,including 168 patients with CAD(CAD group)and 160 normal persons(control group).[Results]Compared with T/T genotype of APOA5,adjusted OR of C/C genotype is 2.45,P=0.01;Adjusted OR of C/C+T/C is 1.72,P=0.02.And C/C genotype appeared more frequently in patients with CAD than normal population.C/C genotype and T/C+C/C genotype might contribute to an increased risk of CAD among Chinese.[Conclusions]ApoA5 gene-1131T→C single nucleotide polymorphism seems to closely associated with CAD in Chinese Han population.The C allele of APOA5 gene-1131T→C single nucleotide polymorphism might be an higher risk factor of CAD among Chinese Han population.

Abstract: [Objective] To identify the inhibitory effects and mechanism of Quercetin on gastric cancer.[Methods] Human gastric cancer cell line SGC7901 cells werer treated with different dose of Quercetin,the growth rate of cancer cells,telomerase activity and apoptosis were evaluated by MTT assay,TRAP-ELISA and TUNEL assay respectively.[Result] Quercetin had a remarkable supression effect on growth of gastric cancer SGC-7901 cells.Anticancer effect of Quercetin was correlation with the supression of telomerse activity.Compared with control groups,significant apoptosis was observed in cancer cells treated with Quercetin.[Conclusion] Quercetin can inhibit proliferation of human gatric cancer cells by inhibiting telomerase activity and induction of apoptosis.

Abstract: [Objective] To investigate the role of chloride channels in cis-diaminedichloroplatinum(cDDP)-induced apoptotic volume decrease and apoptosis in nasopharyngeal carcinoma cells(CNE-2Z).[Methods] cDDP was used to induce CNE-2Z cell apoptosis.Apoptotic cells were detected with the flow cytometry.The time-lapse image acquisition and analysis system was used to detect the volume changes of living cells.[Results] ①An early cell volume decrease was induced by cDDP(5 μmol/L) in CNE-2Z cells.The cell shrunk gradually in the period observed and the volume decreased by 14.2%±2.6% in 6 h.②The chloride channel blocker,5-nitro-2-(3-phenylpropylamino) benzoic acid(NPPB,100 μmol/L) inhibited significantly the cDDPinduced cell volume decrease by 98.6%±1.8%.③NPPB(100 μmol/L) inhibited the cDDP-induced apoptosis by 83.5%±10.4%.The apoptotic rate was decreased from 23.1%±2.5% in the cDDP group to 3.8%±0.5% in the cDDP+NPPB group.[Conclusion] Chloride channels may mediate cDDP-induced CNE-2Z cell apoptosis through inducing apoptotic volume decrease.

Abstract: [Objective] To find an effective medicine in treating diabetic retinopathy(DR).[Methods] DR patients were randomly divided into kakkonein group and control group.We analyzed and compared the hemadynamics and hemorheology indexes of the two groups before and after treatment.[Results] Volume packed cells,whole blood viscosity,plasma viscosity and erythrocyte aggregation index in kakkonein group were lower than those in control group(P0.01),The peak velocity of retinal central artery and eye artery during systolic phase and blood flow rate in final diastolic phase were significantly increased while the resistent index was significantly decreased(P0.01).[Conclusion] Kakkonein is effective for DR.

Abstract: [Objective] To explore the relationship between +276GT polymorphism of adiponectin gene and type 2 diabetic with vascular diseases.[Methods] For the case-control study,polymerase chain reaction-restriction fragment length polymorphism assay(PCR-RFLP)method was used to determine the distribution of genotype frequencies of +276GT polymorphism of the adiponectin gene in 198 cases of type 2 diabetes and 98 normal control subjects.[Results] The frequencies of +276TT homozygotes were significantly increased in control subjects compared with type 2 diabetes(P0.01).There was no significant difference in the distribution of genotype between type 2 diabetes with or without vascular dieases.[Conclusion] The result suggests that the +276GT polymorphism of adiponectin gene is associated with type 2 diabetes in Shandong Han population,those with the +276TT homozygotes have lower risk of type 2 diabetes.The +276GT polymorphism has no obvious association in type 2 diabetes with vascular diseases.

Abstract: [Objective] To study the expression of PLK1 and PCNA in breast carcinoma and its clinical significance.[Methods] The expression of PLK1 and PCNA was detected by S-P immunohistochemistry method in 32 cases of primary breast carcinoma and 16 cases of breast proliferative disease.The expression levels of PLK1 and PCNA in different age,pathological type,tumor size,histological grades,lymph-node metastasis,stage,the status of ER,PR and HER-2 were analyzed.[Results] The expression of PLK1 between primary breast carcinoma and breast proliferative disease was significantly different(P0.05).The expression rate of LPLK1 and PCNA in primary breast was 56.3%(18/32) and 71.9%(23/32) respectively.PLK1 expression had no correlation with age,pathological type,tumor size,lymph node metastasis,stage and HER-2 status(P0.05),but had correlation with differentiation degree,ER and PR(P0.05).PCNA expression had correlation with differentiation degree,ER(P0.05),no correlation with age,pathological type,tumor size,lymph node metastasis.PR and HER-2 status(P0.05).The expression of PLK1 and PCNA was negative or week in breast proliferative disease.The expression of PLK1 in breast cancer was positively correlated with that of PCNA(P0.05).[Conclusion] PLK1 may play an important role in the development and progression of breast cancer and may be a new target for cancer therapy.

Abstract: [Objective] To detect the expression of HER2 and p300/ CBP protein in no-small cell lung cancer(NSCLC) and analyze the relationship between their expression and clinic significance.[Methods] The protein expressions of HER2 and p300/ CBP in 71 NSCLC tissues and 20 normal lung tissues were by immunochemistry.[Results] The overexpression of HER2 and p300/CBP in normal lung tissue were not found,the protein levels of HER2 and p300/CBP in NSCLC were generally detected with which high expression of HER2 was detected as 34%(24/71)and 76% for p300/CBP(54/71).The rate of protein expression in lung adeno-carcinoma was 45.5%(20/44) while expression rate was 19.9%(4/21) in the squamous carcinoma.These differences have statistics meanings(P0.05).The overexpression rate of lymph transfer group(43.2%,19/44) was remarkably higher than the group,which has not been transferred as well(18.5%,5/27).The two groups had conspicuous significance(P0.05).HER2 overexpression correlated with TNM staging,the overexpression of HER2 in TNM Ⅲ+Ⅳ group was significantly higher than that in TNMⅠ+Ⅱ group(P0.05).[Conclusions] HER2 overexpression correlates with pathological type,TNM staging and lymph node metastasis,and is a poor prognostic factor for NSCLC.p300/ CBP overexpression was not correlated with all the pathological parameters statistically.There was no correlation between HER2 and p300/ CBP expression.

Abstract: [Objective] To investigate the significance and mechanisms of expression of p21-activated kinase 1(PAK1) in colorectal carcinoma cells in vitro.[Methods] PAK1 expression was detected in the 8 established cell lines of colorectal carcinoma with different metastasis and malignant potentials by western blot.[Results] The expression of PAK1 was significantly higher in LoVo,COLO320 and SW620 cell lines compared with HCT116,HT29,SW480,SW1116 and LST cells,the expression of PAK1 protein in HT29 and SW480 cell lines was higher than that in SW1116 and LST cells.Furthermore,the expression of PAK1 in SW1116 and LST cell lines was almost lost.[Conclusion] Overexpression of PAK1 protein may be involved in the tumorigenesis and progression of colorectal carcinoma and it is associated closely with the malignant phenotype of colorectal carcinoma.

Abstract: [Objective] To study the change of transforming growth factor-β_1(TGF-β_1) in patients with diabetic nephropathy and it's intervention with valsartan.[Methods] 62 type 2 diabetes patients were divided into two groups according to the level of urinary albumin excretion rate(UAER).The levels of TGF-β_1,blood sugar and urinary renal function were measured in all patients and normal controls.Above markers were measured after valsartan was given.[Results] The levels of UAER and TGF-β_1 in patients with diabetic nephropathy were significantly higher than that in normal controls(P0.01).The level of UAER positively correlated with the level of TGF-β_1 in patients with diabetic nephropathy.After 8 weeks of valsartan therapy,the levels of UAER and TGF-β_1 decreased significantly in patients with diabetic nephropathy.[Conclusion] There is a positive correlation between TGF-β_1 and UAER in patients with diabetic nephropathy.Valsartan can decrease the level of TGF-β_1 and improve the renal function of patients with diabetic nephropathy.

Abstract: [Objective] To investigate the anti-tumor action and the mechanism of the grub extract.[Methods] Korbor method was used to calculate mice acute oral LD50 values(with their 95% confident limits).The grub extract was administered orally to mice,and tumor inhibition action on S180 sarcoma-bearing mice was observed.Superoxide dismutase (SOD) and malondialdehyde(MDA) were detected by colorimetry.[Results] ①Mice acute oral LD50 value(with their 95% confident limits) of the grub extract was 48.73(46.45-51.12) g/kg.②In the group of low,middle and high dosage,the tumor inhibition rates on mice-bearing S180 were 18.64%,39.47% and 53.24% respectively.③The activity of SOD was higher and the content of MDA was lower in the grub extract treated group than those in control group and approach to the levels of control group(P0.01).[Conclusion] The grub extract belongs to low toxicity Chinese medicine.The grub extract has significantly anti-tumor effect,the inhibition is probably realized by antioxidant effect.

Abstract: [Objective] To study effect of cyclosporin A(CSA) on soluble resistance-related calcium-binding protein(Sorcin) gene expression and clinical effect in multiple myeloma(MM) Patients with multidrug resistance(MDR).[Methods] 32 MM patients(MM group) were administrated with combined chemotherapy,and those with multidrug resistance were given CSA at 8-12 mg·kg-1·d-1 24 h before chemotherapy for 3 d,then 4-6 mg·kg-1·d-1 for 2 d.A semi-quantitative reverse transcription,polwnerase chain reaction(RT-PCR) was used to,detect,Sorcin.gene expression rate in MM group and 27 controls(control group).[Results] Sorcin gene expression rate before treatment was 31.25% and 0.30% in MM and control group(P0.05),was 75.00% and 16.67%in MM patients with multidrug resistance and without multidmg resistance(P0.05).Sorcin gene expression rate in MM patients with multidrug resistanc was 75.00% and 62.50% before and after treatment(P0.05).Complete regression and total effective rate was 25,00% 62.50%(P0.05).[Conclusion] CSA can reverse multidrug resistance and enhance clinical effect but the mechanism has no correlaton with.Sorcin gene expression.

Abstract: [Objective] To explore proteins from Nidus Vespae(NVP) inducing apoptosis of K562 cells in vitro and its mechanism.[Methods] Protein ingredients were extracted from NVP by various means including centrifuge,gel chromatography and dialysis.K562 cells were cultured with NVP1-4.Morphological changes of apoptotic cells were observed via an inverted phase contrast microscope and by Hoechst 33258 stain via a fluorescent microscope(FM).Expressions of caspase-3 was examined by immunohistochemistry.[Results] K562 cells growed more slowly and were decreased.Particles in cytoplasm were increased and there were many cacuoles and fragments cytoplasm.Fluorescence staining enhanced significantly,karyotin showed dense lumps or thick particles.Caspase-3 expression was increased significantly.Compared with blank control group NVP groups all had significant differences(P0.05).[Conclusion] NVP can markedly inhibit the proliferation of K562 cells.The mechanism may be related to upregulation and activation of caspase-3.

Abstract: [Objective] To establish genescan technique of diversity gene of T cell antigen receptor(TCR) complementary-determining region 3(CDR3) for assaying clonality of human TCR αβ T cell.[Methods] We took Jurkat cell and Raji cell as type strain and negative control respectively,and amplified 34 TCR α chain variable gene(TCR Vα) and TCR 26 β chain variable gene(TCR Vβ) of 5 healthy individuals peripheral blood T cell by RT-PCR.Productions of RT-PCR were assayed by genescan technique to determine clonality of TCR αβ T cell.[Results] Only mRNA genes of TCR Vα1.1 and TCR Vβ8 were found in Jurkat cells,no TCR Vα and TCR Vβ ge ne was found in Raji cells.Jurkat cell was monoclonal T cell.All the TCR Vα and TCR Vβ genes were expressed in 5 healthy individuals peripheral blood T cell.T cell were multiclonal.[Conclusion] Genescan technique of diversity gene of TCR CDR3 was stable and precise method for assaying clonality of human TCR αβ T cell.

Abstract: [Objective]To screen bio-marker molecules from the serum of ovarian and laryngeal cancer patients re- spectively via surface-enhanced laser desorption/ionization time-of-flight mass spectrometry(SEIDI-TOF-MS).And to by e- valuate the feasibility of the method on tumor therapeutic effect monitoring comparing the expression of the biomarkers before and after the clinical intervention.[Methods]Serum samples from 20 cases of ovarian cancer patients before and after chemotherapy and 51 cases of laryngeal carcinoma patients before and after operation were analyzed by SEIDI-TOF-MS pro- tein chip.The statistic analysis was done by using Biomarker Wizard software.[Results]In ovarian cancer samples,pro- tein peak 4475(m/z)was found increased in 14/20 patients after the chemotherapy.The protein peak 13741(m/z)was highly expressed in all preoperative patient samples and decreased in 21/51 cases postoperatively.Among this 21 cases, three peaks(13741,5192 and 15100 m/z)were found dramatically decreased in 11 patients after operation(P0.01). [Conclusions]SEIDI-TOF-MS can sensitively detect the change of a protein peak between either pre- and post-operative or pre- and post-chemotherapy,SELDI proteomics technology may have a better foreground on the monitoring of tumor thera- peutic effect for its properties of minor injuries,high sensitivity and small sample volume.

Abstract: [Objective] To investigate the relationship between genetic polymorphisms of MDR-1 and XPG and response rate of platinum-based chemotherapy on non-small-cell lung cancer(NSCLC).[Methods] 61 patients with advanced NSCLC were routinely treated with platinum-based chemotherapy,and clinical response was evaluated after 2~3 cycles.XPG and MDR-1 genotypes were determined by PCR-RFLP Compared the effects of different genotypes on chemotherapy res ponsiveness.[Results]The response rate to chemotherapy in patients with the allele MDR1-3435 C/C was significantly higher than that with the T/C or T/T genotype(59.3% vs 26.5%;OR=0.442,95%CI=0.133~1.467,P 0.05);The response rate to the chemotherapy in patients with MDR-1 2677 T/T or G/T genotype was significantly lower than that in patients with other genotypes(13.0% vs 57.9%;OR=0.384,95%CI=0.187~0.789,P 0.01);While there was no statistically significant correlation between the XPG polymorphisms and response rate to chemotherapy.[Conclusion] Genetic polymorphisms of MDR-1 C3435T、MDR-1 G2677A/T can degrade the response rate in NSCLC patients treated with platinum-based chemotherapy.There is no association between the polymorphisms of XPG C3507G and response to platinum-based chemotherapy.

Abstract: [Objective] To investigate the relationship between degeneration of cartilage and the tumor necrosis factor α(TNF-α),transforming growth factor β(TGF-β),interleukin 6(1L-6),IL-9 in osteoarthritis(OA).[Methods] The histological changes of cartilages by hematoxyllin-eosin staining and immunohisto-chemical.The expression of TNF-α,TGF-β,IL-6 and IL-9 were studied in 20 OA cases and 2 normal controls.[Results] The OA cartilage showed fibroplasias.The quantity of chondrocyte increased and the clustered and hypertrophic cells came into being.Little TNF-α and IL-6 was observed in normal cartilage,while their expressions increased in degenerated cartilage of OA(P 0.01).[Conclusions] The abnormal expression of TNFα,IL-6 and TGF-β,IL-9 is likely to be the pathogenesis of cartilage degeneration.

Abstract: [Objective] To study the role of promoter hypermethylation of APC gene tumorigenesis and development in epithelial ovarian carcinoma.[Methods] Promoter hypermethylation of APC was detected by methylation-specific PCR in 63 epithelial ovarian cancer patients,41 metastatic tissues of pelvic and abdomen cavity,10 adjacent non-cancerous ovarian tissues and 20 normal ovarian tissues.[Results] Promoter hypermethylation of APC was detected in ovarian cancer tissues and metastatic sites,the rates were 22.2% and 22.0% respectively,higher than that in normal ovarian tissues significantly(P0.05).No statistical difference was found between different tumor stages,histological types and different differentiated grades in the frequency of promoter hypermethylation of APC(P0.05).[Conclusion] Promoter hypermethylation of APC plays an important role in the course of tumorigenesis and development in epithelial ovarian carcinoma.

Abstract: [Objective] To investigate the effect of hepatitis C virus non-structural protein 4B(HCV NS4B)on wild-type p53 gene expression and apoptosis in hepatic cells.[Methods] The empty vector PCXN2,HCV NS4B,recombinant plasmid PCXN2-NS4B and wild-type p53 expression plasmid were transfected or co-transfected into Chang liver cells.Screening was performed with G418.Plasmid transfected into hepatic cells successfully was detected by reverse transcription PCR.The expressions of p53 gene in PCXN2 group,NS4B group,p53 group,NS4B-p53 group were detected by using in-situ hybridization.TUNEL(flow cytometry) was used for assessing the apoptosis rates.[Results] Compared with the PCXN2 group,p53 mRNA expression was not promoted or inhibited in NS4B group(P0.05);Compared with p53 group,p53 mRNA expression in normal liver cells of NS4B-p53 group was degraded(P0.01);The cell apoptosis rates in PCXN2 group,NS4B group,p53 group,NS4B-p53 group were respectively(17.02±1.24)%,(11.94±2.24)%,(25.84±3.49)%,(18.34±1.55)%.[Conclusion] HCV NS4B may inhibit p53 gene expression and liver cells apoptosis.HCV NS4B may inhibit cell apoptosis by inhibiting p53 expression.

Abstract: [Objective] To investigate the influence of insulin resistance(IR) and hyperinsulinemia(HIS) on ultrasonocardiogram changes of cardiac damage in essential hypertension(EH) patients.[Methods] The oral glucose tolerance test(OGTT)was carried out in 60 EH patients,Insulin,C peptide releasing and ultrasonocardiogram were assessed at the same time.[Results]No significant difference was found in blood glucose on every phase between HIS group and control group(P0.05),but the levels of serum insulin and C peptide in HIS group were higher than those of control group(P0.01).Insulin action index in HIS group was obviously lower than that in control group(P0.01).Cordis morphology changes and interventricular mass index,interventricular septal thickness in the two group were all significantly different(P0.05).A velocity and A/E in HIS group were obviously higher than those of control group(P0.05).[Conclusion] EH patients with HIS have IR.IR and HIS promote the genesis and development of EH left ventricular hypertrophy and the damage of EH cardiac diastolic function.

Abstract: [Objective] To investigate generation of prokaryotic expressions of antitumor associated g1ycoprotein-72(TAG-72)scFv,and to detect TAG-72 expression in human primary gallbladder carcinomas(PGC).[Methods] Anti-TAG-72 scFv was prokaryotic-expressed and proved to be 31 kDa fuse-protein by SDS-PAGE and Western blot.Exporesstion of TAG-72 antigen was determined in gallbladder tumor by immunohisto-chemistry.[Result] The positive rates of TAG-72 in 47 PGCs and 15 gallbladder adenomahyperplastics were 59.6%and6.7%,respectively.It shows that TAG-72 expression in PGC was significantly higher than those ingallbladder adenoma-hyperplastics(P 0.05);and that expression in well differentiated PGC(48.3%) was significantly lower than those in moderately,poorly differentiated PGC(48.3%,P 0.05).TAG-72 was more highly expressed in smaller PGC(26.3%)than larger one(82.1%,P 0.05),but no statistical difference between various PGC clinical stages.[Conclusion] We generated anti-TAG-72 scFv by prokaryotic expression and detected TAG-72 expression in human gallbladder tumors,it suggests that TAG-72 could be used as gallbladder tumor′s maker in clinical applications.

Abstract: [Objective] To study the relationship between methylation and expression,and detect the difference of methylation status in promoter of Zonula occludens 1(ZO-1) gene between human acute leukemia cell lines and normal bone marrows.[Methods] The expression of ZO-1 gene in three leukemia cell lines and the normal bone marrows was studied with reverse transcription PCR.The methylation pattern in promoter region of ZO-1 gene was detected with methylation specific PCR and the HL60 cell line was treated with demethylation drugs to investigate the relationship between methylation and expression of ZO-1 gene.[Results] ZO-1 gene was expressed in normal bone marrow,but not in leukemia cell lines.The promoter region of ZO-1 gene was unmethylated in normal bone marrow but it was completely methylated in leukemia cell lines.After treated with the demethylation drugs,the methylation pattern in promoter region of ZO-1 in HL60 was dimethylated and this results in the expression of ZO-1 gene again.[Conclusions] The hypermethylation of the promoter region of ZO-1 gene in acute leukemia cell results in expression silence of ZO-1.ZO-1gene may be associated with a new pathogenesis of leukemia.

Abstract: [Objective] To study the expression of multidrug resistance gene(mdr1) and its clinical significance.[Methlods] A semi-quantitative reverse transcription polymerase chain reaction(RTPCR)was used to investigate the transcription levels of the human sorcin gene detected by Northern blot in 42 ALL patients and 27 controls,and analyzed the clinical significance of mdr1 gene.[Results] Mdr1 gene expression was significantly higher in ALL patients than that in normal controls(P0.01),and higher in relapsed/refractory ALL patients than that in those newly diagnosed or in complete remission.Mdr1 gene expression was significantly lower in non-resistant patients than that in resistant ones(P0.01).CR rates of non-resistant patients was higher than of resistant ones(P0.001).[Conclusion] Mdr1 gene overexpression is significantly associated with clinical multidrug resistance and prognosis,and it is one of the indicators for multidrug resistance and prognosis of ALL patients.
