Anti-Bacterial protein extracted from Lactobacillus plantarum (VITSE07) targeting food borne pathogens

Abstract: This study deals with optimization and application of the proteinaceous toxin bacteriocin produced from Lactobacillus plantarum VITSE07. The bacterial strain culture L. plantarum VITSE07used in this research was isolated from the curd sample, a fermented milk product and characterized.The16SrRNA sequence of VITES07strain has been submitted to EMBL database. It showed effective Bacteriocin production against target pathogens Listeria monocytogenes(MTCC65), Staphylococcus aureus(NCIM 20), E.coli (NCIM 2931), Pseudomonas spand Salmonella typhi (NHRC). It showed high stability at 30°C in temperature, pH 3 and 5% of inoculum concentration and the protein concentration was estimated by Lowry’s method. Bacteriocin was purified by ammonium sulfate precipitation followed by dialysis, gel filtration chromatography and High performanace liquid chromatography. The molecular weight of bacteriocin was found to be 29 kda using SDS PAGE. Finaally application study carried out for crude as well as purified bacteriocin to check for biopreservation on two varieties of sprouts Macrotyloma uniflorum(Horse gram) and Vigna radiate (Green gram). These could be preserved for more than 20 days at 4°C respectively.
