The objective of this study is to isolate, identify and evaluate the effect of Klebsiella pnemoniae in relation to the catfish, by isolating the microorganisms present on the catfish skin and intestine. To find out if Klebsiella pneumoniae bacteria isolate can survive in the fish (immunity of fish) and observation of the public health hazard that bacteria i.e. test organism and natural flora exposes the people to, samples of catfish were analyzed using standard techniques. The physicochemical analysis showed that there were changes in the pH, the temperature and the dissolved oxygen of the water samples. The distributions of the bacteria species isolated from the catfish intestine and on the skin showed that Klebsiella pneumoniae [12(25.0%)] was the most predominant bacteria isolates. This was followed by Enterococcus feacalis [9(18.6%)], Staphylococcus aureus [7(14.6%)], Escherichia coli [6(12.5%)], Citrobacter ferundii [5(10.4%)], Pseudomonas aeruginosa [5(10.4%)], and Bacillus subtilis [4(8.3%)]. It also showed that Enterococcus feacalis and Citrobacter ferundii were absent in all the skin samples of the catfish but present in all the intestines. Klebsiella pneumoniae was present on all the skin and intestines of the catfish samples. Escherichia coli were present in all the skin and intestine except for skin of the catfish samples in Experiment 1. Bacillus subtilis was only present in the skin of the catfishes used as control. Staphylococcus aureus was absent in all the intestine of the catfishes samples but present in their skins. Pseudomonas aeruginosa was absent in all the samples except for the intestines of the catfishes used as control and those in experiment 2. Generally, Klebsiella pneumoniae inoculated into catfish Clarias gariepinus presented to be virulent in the fishes leading to shedding of skin patches and fading of colour on skin from black to faint black. The findings of this study showed that Klebsiella pneumoniae may cause an infection in catfish and can act as a vector of human pathogen. Furthermore, other bacteria were isolated which may be resident flora of the catfish. Xanthomonas oryzae pv.oryzae, the causal or-ganism of bacterial blight of rice which produces leaf blight as well as kresek (wilt) symptoms in plants were tested for indole, auxin production in culture supplemented withl-tryptophan. On the basis of indoleacetic acid (IAA) production the isolates were grouped into IAA-positive and IAA-negative. Out of 17 isolates, 11 were IAA-positive while 6 were IAA-negative. The isolates metabolized tryptophan through two different routes and the isolates vary in the pathway of tryptophan utilization. The IAA-positive isolates converted tryptophan to IAA as the end product, whereas the IAA-negative isolates formed anthranilate as an intermediate metabolite and finally produced pyrocatecholvia the kynurenine pathway. Quantification of tryptophan metabolism revealed that the maximum production of IAA and pyrocatechol in culture occurred during 2-d incubation at 30±2°C.

An evaluation and analysis of nearshore water quality data has been undertaken. These data have been collected along the Canadian shoreline of the Great Lakes between 1967 and 1973 by the Ontario Ministry of the Environment (O.M.E.). The nearshore study fits within the framework of a much larger ongoing program at Queen's University for the purpose of storage, management and analysis of hydrometric, water quality and physical data undertaken for the International Joint Commission's (I.J.C.) Pollution from Land Use Activities Reference Group (PLUARG). The recognition, by both Canada and the United States, of the continued deterioration of Great Lakes water quality and the heavy and increasing use of the nearshore waters, often for incompatible purposes (for example recreation and waste disposal) makes a thorough study of existing nearshore water quality data necessary. The single most difficult problem in dealing with this data set is its high degree of spatial and temporal variability; therefore, considerable time and effort was expended in obtaining an appreciation of its complexities. This is achieved with detailed summaries of station and parameter sampling frequencies. Parameters which meet specific sampling frequency criteria are identified as being suitable for statistical analysis. However, only eight parameters, generally applicable to a description of the health of the nearshore zone, are considered in this report. The complex and transitory nature of the limnologic processes affecting nearshore water quality, coupled with the inherent temporal-spatial variability of the data set, suggest the need for data aggregation (illustrated below). The intent of data aggregation and the underlying assumption of the statistical summary of nearshore water quality data is that aggregation homogenizes the effects of process variables as well as providing data subsets containing relatively large data populations which, in turn, enhance statistical confidence. Objective: To design and develop an ultraviolet germicidal irradiation (UVGI) device for controlling airborne bacteria in the workplace of hospitals. Material and Method: The constructed UVGI device was a chamber, consisting of a return air grille, an air supply grille, two 30-watt ultraviolet lamps and a cross flow fan inside the chamber. The efficiency of the UVGI device was tested at three different flow rates of 15.7, 31.6 and 46.1 m 3 /min, respectively by collecting bacteria samples before and after operating the device for 30, 60, 90 and 120 minutes. Results: The efficiency of the UVGI device at flow rates of 15.7, 31.6 and 46.1 m 3 /min was 60.6, 92.8 and 80.8%, respectively and the bacteria concentration was reduced to less than 100 cfu/m 3 , except at the flow rate of 15.7 m 3 /min. The application of the UVGI device in the specimen-received room showed that the concentration of bacteria was reduced to less than 100 cfu/m 3 after 180 minutes. Conclusion: The constructed UVGI device was effective to reduce airborne bacteria in workplace of hospital.
