Validated prediction of clinical outcome in sarcomas and multiple types of cancer on the basis of a gene expression signature related to genome complexity
Frédéric Chibon,Pauline Lagarde,Sébastien Salas,Gaëlle Pérot,Véronique Brouste,Franck Tirode,Carlo Lucchesi,Aurélien de Reyniès,Audrey Kauffmann,Binh Bui,Philippe Terrier,Sylvie Bonvalot,Axel Le Cesne,Dominique Vince-Ranchère,Jean-Yves Blay,F. Collin,Louis Guillou,Agnès Leroux,Jean-Michel Coindre,Alain Aurias +19 more
TL;DR: A prognostic gene expression signature, complexity index in sarcomas (CINSARC), composed of 67 genes related to mitosis and chromosome management, is established, which predicts metastasis outcome and predicts outcome for gastrointestinal stromal tumors, breast carcinomas and lymphomas.
Journal ArticleDOI
HMGA2 is the partner of MDM2 in well-differentiated and dedifferentiated liposarcomas whereas CDK4 belongs to a distinct inconsistent amplicon.
Antoine Italiano,Laurence Bianchini,Frédérique Keslair,S. Bonnafous,Nathalie Cardot-Leccia,Jean-Michel Coindre,Jean-Marc Dumollard,Paul Hofman,Agnès Leroux,Claire Mainguené,Isabelle Peyrottes,Dominique Ranchère-Vince,Philippe Terrier,Albert Tran,Philippe Gual,Florence Pedeutour +15 more
TL;DR: The 12q13‐15 amplicon which contains MDM2 and CDK4 in well‐differentiated and dedifferentiated liposarcomas (WDLPS/DDLPS) was investigated using fluorescence in situ hybridization analysis and found a frequent amplification and overexpression of YEATS4, an oncogene that inactivates P53, suggesting that YEATs4 might play an important role together with MDM 2 in WDL PS/DD
Journal ArticleDOI
High frequency of β -catenin heterozygous mutations in extra-abdominal fibromatosis: a potential molecular tool for disease management
Julien Domont,Sébastien Salas,Ludovic Lacroix,Véronique Brouste,Patrick Saulnier,P. Terrier,Dominique Ranchère,Agnès Neuville,Agnès Leroux,Louis Guillou,Raf Sciot,F. Collin,Armelle Dufresne,J.-Y. Blay,A. Le Cesne,Coindre Jm,S. Bonvalot,Jean Bénard +17 more
TL;DR: Wild-type β-catenin seems to be an interesting prognostic marker that might be useful in the therapeutic management of extra-abdominal fibromatosis from a large multicentric retrospective study.
Journal ArticleDOI
Clinical and Biological Significance of CDK4 Amplification in Well-Differentiated and Dedifferentiated Liposarcomas
Antoine Italiano,Laurence Bianchini,Elisabet Gjernes,Frédérique Keslair,Dominique Ranchère-Vince,Jean-Marc Dumollard,Juliette Haudebourg,Agnès Leroux,Claire Mainguené,Philippe Terrier,Frédéric Chibon,Jean-Michel Coindre,Florence Pedeutour +12 more
TL;DR: It is found that MDM2+/CDK4- WDLPS/DDLPS represent a distinct clinical subgroup with favorable prognostic features, including low-grade lipoma-like histology, peripheral location, and lower rate of recurrence.
Journal ArticleDOI
Molecular characterization by array comparative genomic hybridization and DNA sequencing of 194 desmoid tumors.
Sébastien Salas,Frédéric Chibon,Tetsuro Noguchi,Philippe Terrier,Dominique Ranchère-Vince,Pauline Lagarde,Jean Bénard,Sébastien Forget,Camille Blanchard,Julien Domont,Sylvie Bonvalot,Louis Guillou,Agnès Leroux,Agnes Mechine-Neuville,Patrick Schöffski,Marik Laë,F. Collin,Olivier Verola,Amelie Carbonnelle,Laure Vescovo,Binh Bui,Véronique Brouste,Hagay Sobol,Alain Aurias,Jean-Michel Coindre +24 more
TL;DR: A nonrandom occurrence of trisomy 8 associated with an increased risk of recurrence is revealed and Alterations of APC, including loss of the entire locus, and CTNNB1 mutation could explain the tumorigenesis in 89% of sporadic desmoid tumors and desmoids tumors occurring in the context of Gardner's syndrome.
