Journal ArticleDOI
Delivery of ENaC siRNA to epithelial cells mediated by a targeted nanocomplex: a therapeutic strategy for cystic fibrosis.
Maria Manunta,Aristides D. Tagalakis,Martin Attwood,Martin Attwood,Ahmad Aldossary,Josephine Barnes,Mustafa M. Munye,Alexander Weng,Alexander Weng,Robin J. McAnulty,Stephen L. Hart +10 more
TL;DR: Findings show that LPRs can modulate the activity of ENaC and this approach might be promising as co-adjuvant therapy for cystic fibrosis.
Journal ArticleDOI
Effective silencing of ENaC by siRNA delivered with epithelial-targeted nanocomplexes in human cystic fibrosis cells and in mouse lung.
Aristides D. Tagalakis,Mustafa M. Munye,Rositsa Ivanova,Hanpeng Chen,Claire Smith,Ahmad Aldossary,Luca Rosa,Dale Moulding,Josephine Barnes,Konstantinos N. Kafetzis,Stuart A. Jones,Deborah L. Baines,Guy W. J. Moss,Christopher O'Callaghan,Robin J. McAnulty,Stephen L. Hart +15 more
TL;DR: Nanoparticle-mediated delivery of ENaCsiRNA to ALI cultures corrected aspects of the mucociliary defect in human CF cells and offers effective delivery and silencing in vivo.
Journal ArticleDOI
Peptide and nucleic acid-directed self-assembly of cationic nanovehicles through giant unilamellar vesicle modification: Targetable nanocomplexes for in vivo nucleic acid delivery.
Aristides D. Tagalakis,Ruhina Maeshima,Cynthia Yu-Wai-Man,Jinhong Meng,Farhatullah Syed,Lin-Ping Wu,Ahmad Aldossary,David McCarthy,Seyed Moein Moghimi,Stephen L. Hart +9 more
TL;DR: Novel nanovesicle complexes derived from giant unilamellar vesicle precursors comprising self-assembling mixtures of the nanovesicles, plasmid DNA or siRNA and targeting peptide ligands allow for the specific targeted enhancement of nucleic acid delivery with improved biosafety over liposomal formulations and represent a promising tool to improve the arsenal of safe, non-viral vectors to deliver therapeutic cargos in a variety of disorders.
Journal ArticleDOI
Fast-Dissolving Nifedipine and Atorvastatin Calcium Electrospun Nanofibers as a Potential Buccal Delivery System
Hassa A. Alshaya,Ahmed Alfahad,Fatemah M. Alsulaihem,Alhassan H. Aodah,Abdullah Alshehri,Fahad A. Almughem,Haya A. Alfassam,Ahmad Aldossary,Abdulrahman A. Halwani,Haitham Bukhary,Moutaz Badr,Salam Massadeh,Manal Alaamery,Essam A. Tawfik +13 more
TL;DR: The development of drug-loaded nanofibers containing nifedipine and atorvastatin calcium can be a potential buccal delivery system for geriatric patients' convenience.
Journal ArticleDOI
Integrin‐Targeted, Short Interfering RNA Nanocomplexes for Neuroblastoma Tumor‐Specific Delivery Achieve MYCN Silencing with Improved Survival
Aristides D. Tagalakis,Vignesh Jayarajan,Ruhina Maeshima,Kin H. Ho,Farhatullah Syed,Lin-Ping Wu,Ahmad Aldossary,Mustafa M. Munye,Talisa Mistry,Olumide Ogunbiyi,Arturo Sala,Joseph F. Standing,Seyed Moein Moghimi,Andrew W. Stoker,Stephen L. Hart +14 more
TL;DR: RTN formulations can achieve specific tumor‐targeting, with minimal clearance by the liver and so enable delivery of tumor-targeted siRNA therapeutics, and are shown to be RGD peptide‐dependent indicating integrin‐mediated uptake.
