Journal ArticleDOI
A broad-host-range Flp-FRT recombination system for site-specific excision of chromosomally-located DNA sequences: application for isolation of unmarked Pseudomonas aeruginosa mutants.
TL;DR: An improved method for gene replacement in Pseudomonas aeruginosa was developed and a cassette was constructed that contains a GmR selectable marker next to the green fluorescent protein structural gene, with both markers being flanked by Flp recombinase target (FRT) sites.
Journal ArticleDOI
Characterization of a pseudomonas aeruginosa fatty acid biosynthetic gene cluster : purification of acyl carrier protein (acp) and malonyl-coenzyme a :acp transacylase (fabd)
TL;DR: This work purified FabD as a hexahistidine fusion protein (H6-FabD) and ACP in its native form via an ACP-intein-chitin binding domain fusion protein, using a novel expression and purification scheme that should be applicable to ACP from other bacteria.
Journal ArticleDOI
High-plex imaging of RNA and proteins at subcellular resolution in fixed tissue by spatial molecular imaging.

Beechem +37 more
TL;DR: S spatial molecular imaging is described, a system that measures RNAs and proteins in intact biological samples at subcellular resolution by performing multiple cycles of nucleic acid hybridization of fluorescent molecular barcodes, and has high sensitivity and very low error rate.
Journal ArticleDOI
Abstract 1364: Spatially resolved T-cell receptor profiling elucidates relationships between TCR diversity, immune infiltration, and cancer-associated pathways
Michelle A. Kriner,Katrina van Raay,Jason Reeves,Kit Fuhrman,Andrew Klock,Alecksandr J. Kutchma,Erin Piazza,Joseph M. Beechem +7 more
TL;DR: The ability to link the spatial context of TCR segment expression to activation of malignant signaling cascades as well as non-cancerous T-cell response to the presence of other immune cells and cancer-associated signaling is demonstrated.
