Journal ArticleDOI
Molecular and Histopathological Changes Associated with Keratoconus
TL;DR: The histopathological features of different layers of cornea and the differentially expressed proteins in the KC-affected cornea are summarized and discussed to help emphasize the major molecular defects in KC and identify additional research areas related to KC.
Journal ArticleDOI
Differential Expression of Coding and Long Noncoding RNAs in Keratoconus-affected Corneas
Mariam Lofty Khaled,Yelena Bykhovskaya,Sarah E.R. Yablonski,Hanzhou Li,Michelle Drewry,Inas F. Aboobakar,Amy Estes,Xiaoyi Gao,W. Daniel Stamer,Hongyan Xu,R. Rand Allingham,Michael A. Hauser,Yaron S. Rabinowitz,Yutao Liu +13 more
TL;DR: This study highlighted the potential roles of several genes (CTGF, SFRP1, AQP5, lnc-WNT4-2-2:1, and lNC-ALDH3A2- 2:1) and pathways (TGF-β, WNT signaling, and PI3K/AKT pathways) in KC pathogenesis.
Journal ArticleDOI
PPIP5K2 and PCSK1 are Candidate Genetic Contributors to Familial Keratoconus.
Mariam Lofty Khaled,Mariam Lofty Khaled,Yelena Bykhovskaya,Chunfang Gu,Alice Liu,Michelle Drewry,Zhong Chen,Barbara A. Mysona,Barbara A. Mysona,Emily J. Parker,Ryan P. McNabb,Hongfang Yu,Xiaowen Lu,Jing Wang,Xiaohui Li,Abdulrahman Al-Muammar,Jerome I. Rotter,Louise F. Porter,Amy Estes,Amy Estes,Mitchell A. Watsky,Mitchell A. Watsky,Sylvia B. Smith,Sylvia B. Smith,Hongyan Xu,Khaled K. Abu-Amero,Anthony N. Kuo,Stephen B. Shears,Yaron S. Rabinowitz,Yutao Liu,Yutao Liu +30 more
TL;DR: Using in vitro cellular model and in vivo gene-trap mouse model, critical evidence is found to support the role of PPIP5K2 in normal corneal function and KC pathogenesis and for the first time,Corneal tomography and pachymetry mapping are integrated into characterization of mouse cornea phenotypes which could be widely implemented in basic and translational research for KC diagnosis and therapy.
Journal ArticleDOI
Dioleoylphosphatidylglycerol Accelerates Corneal Epithelial Wound Healing.
Wendy B. Bollag,Lawrence O. Olala,Lawrence O. Olala,Ding Xie,Xiaowen Lu,Haixia Qin,Haixia Qin,Vivek Choudhary,Vivek Choudhary,Rachana Patel,David Bogorad,Amy Estes,Mitchell A. Watsky +12 more
TL;DR: The results indicate the importance of the AQP3/PLD2/PG signaling pathway in corneal epithelial cells and suggest the possibility of developing DOPG as a pharmacologic therapy to enhanceCorneal wound healing in patients.
Journal ArticleDOI
Comparison of Different Mass Spectrometry Workflows for the Proteomic Analysis of Tear Fluid
Garrett C. Jones,Tae Jin Lee,Joshua Glass,Grace Rountree,Lane Ulrich,Amy Estes,Mary E Sezer,Wenbo Zhi,Shrutie Sharma,Ashok Sharma +9 more
TL;DR: A method of in-strip protein digestion that identified >3000 proteins in human tear samples from 11 healthy subjects is established, which offers a significant improvement in the number of proteins identified compared to previously reported methods without pooling samples.
