Journal ArticleDOI
Cell death induced by dexamethasone in lymphoid leukemia is mediated through initiation of autophagy
Edward Laane,K. Pokrovskaja Tamm,Eva Buentke,Keisuke Ito,P. Khahariza,J. Oscarsson,Martin Corcoran,Ann-Charlotte Björklund,Kjell Hultenby,Johan Lundin,M. Heyman,Stefan Söderhäll,Joanna Mazur,Anna Porwit,Pier Paolo Pandolfi,Boris Zhivotovsky,Theocharis Panaretakis,Dan Grandér +17 more
TL;DR: It is shown that dexamethasone induced autophagy upstream of apoptosis in acute lymphoblastic leukemia cells, and inhibited apoptosis by siRNA-mediated repression of Beclin 1 expression, showing an important role of Autophagy in dexamthasone-induced cell death.
Journal ArticleDOI
Acute hypoxia induces apoptosis of pancreatic β-cell by activation of the unfolded protein response and upregulation of CHOP.
Xiaowei Zheng,X Wang,Z Ma,V Gupta Sunkari,Ileana Ruxandra Botusan,T Takeda,Ann-Charlotte Björklund,M Inoue,S.-B. Catrina,Kerstin Brismar,Lorenz Poellinger,Lorenz Poellinger,Teresa Pereira +12 more
TL;DR: It is observed that pancreatic islets of type 2 db/db diabetic mice were more hypoxic than their counterpart in normoglycemic animals, which indicates that hypoxia-mediated apoptosis may occur in type 2 diabetes.
Journal ArticleDOI
Targeting of distinct signaling cascades and cancer-associated fibroblasts define the efficacy of Sorafenib against prostate cancer cells.
Pedram Kharaziha,Patricia Q. Rodriguez,Qiao Li,Helene Rundqvist,Ann-Charlotte Björklund,Martin Augsten,Anders Ullén,Lars Egevad,Peter Wiklund,Sten Nilsson,Guido Kroemer,Dan Grandér,Theocharis Panaretakis +12 more
TL;DR: The differential tyrosine kinase profile of prostate cancer cells defines the cytotoxic efficacy of sorafenib and this profile is modulated by CAFs to promote resistance.
Journal ArticleDOI
Glucocorticoid-induced cell death is mediated through reduced glucose metabolism in lymphoid leukemia cells
Eva Buentke,Anders Nordström,H Lin,Ann-Charlotte Björklund,Edward Laane,Masako Harada,Li Lu,Tetyana Tegnebratt,Sharon Stone-Elander,M. Heyman,Stefan Söderhäll,Anna Porwit,CG Östenson,M Shoshan,K. Pokrovskaja Tamm,Dan Grandér +15 more
TL;DR: The data show that GC treatment blocks glucose uptake by leukemic cells leading to inhibition of glycolysis and that these effects play an important role in the induction of cell death by these drugs.
Journal ArticleDOI
Metabolic reprogramming of acute lymphoblastic leukemia cells in response to glucocorticoid treatment.
Matheus Dyczynski,Mattias Vesterlund,Ann-Charlotte Björklund,Vasilios Zachariadis,Jerry Janssen,Hector Gallart-Ayala,Evangelia Daskalaki,Craig E. Wheelock,Janne Lehtiö,Dan Grandér,Katja Pokrovskaja Tamm,Roland Nilsson,Roland Nilsson +12 more
TL;DR: The data suggest that glutamine synthesis affects autophagy and possibly onset of cell death in response to GCs, which should be further explored to understand mechanism of action and possible sources of resistance.
