Delineating the role of cooperativity in the design of potent PROTACs for BTK
Adelajda Zorba,Chuong Nguyen,Yingrong Xu,Jeremy T. Starr,Kris A. Borzilleri,James F. Smith,Hongyao Zhu,Kathleen A. Farley,WeiDong Ding,James Schiemer,Xidong Feng,Jeanne S. Chang,Daniel P. Uccello,Jennifer A. Young,Carmen N. Garcia-Irrizary,Lara C. Czabaniuk,Brandon P. Schuff,Robert M. Oliver,Justin I. Montgomery,Matthew Merrill Hayward,Jotham Wadsworth Coe,Jinshan Chen,Mark Niosi,Suman Luthra,Jaymin C. Shah,Ayman El-Kattan,Xiayang Qiu,Graham M. West,Mark C. Noe,Veerabahu Shanmugasundaram,Adam M. Gilbert,Matthew Frank Brown,Matthew F. Calabrese +32 more
TL;DR: It is shown that within a Bruton’s tyrosine kinase/cereblon PROTAC system, potent knockdown correlates with alleviation of steric clashes in the absence of thermodynamic cooperativity, which broadens the scope of PROTAC applications and affects fundamental design criteria across the field.
Journal ArticleDOI
Development of a new permeability assay using low-efflux MDCKII cells
Li Di,Carrie Whitney-Pickett,John P. Umland,Hui Zhang,Xun Zhang,David F. Gebhard,Yurong Lai,James Federico,Ralph E. Davidson,Russ Smith,Eric L. Reyner,Caroline A. Lee,Bo Feng,Charles J. Rotter,Manthena V.S. Varma,Sarah Kempshall,Katherine S. Fenner,Ayman El-Kattan,Theodore E. Liston,Matthew D. Troutman +19 more
TL;DR: A new cell line, MDCKII-LE (low efflux), has been developed by selecting a subpopulation of low-efflux cells from MDCkII-WT using an iterative fluorescence-activated cell sorting technique with calcein-AM as a Pgp and efflux substrate.
Journal ArticleDOI
Physicochemical Space for Optimum Oral Bioavailability: Contribution of Human Intestinal Absorption and First-Pass Elimination
Manthena V.S. Varma,R. Scott Obach,Charles J. Rotter,Howard Miller,George Chang,Stefanus J. Steyn,Ayman El-Kattan,Matthew D. Troutman +7 more
TL;DR: In conclusion, physicochemical properties influence bioavailability with typically opposing effects on Fa and first-pass elimination, which may provide a rational judgment on the physicochemical space to optimize oral bioavailability.
Journal ArticleDOI
Mechanistic pharmacokinetic modeling for the prediction of transporter-mediated disposition in humans from sandwich culture human hepatocyte data.
Hannah M. Jones,Hugh A. Barton,Yurong Lai,Yi An Bi,Emi Kimoto,Sarah Kempshall,Sonya C. Tate,Ayman El-Kattan,J. Brian Houston,Aleksandra Galetin,Katherine S. Fenner +10 more
TL;DR: This study illustrates the mechanistic and model-driven application of in vitro uptake and efflux data for human PK prediction for OATP substrates and shows the ability to capture the multiphasic plasma concentration-time profiles for such compounds using only preclinical data.
Journal ArticleDOI
Predicting Clearance Mechanism in Drug Discovery: Extended Clearance Classification System (ECCS).
TL;DR: The proposed ECCS framework is valuable in early prediction of clearance mechanism and can aid in choosing the right preclinical tool kit and strategy for optimizing drug exposure and evaluating clinical risk of pharmacokinetic variability caused by drug interactions and pharmacogenomics.
