Journal ArticleDOI
Risk stratification of childhood medulloblastoma in the molecular era: the current consensus
Vijay Ramaswamy,Marc Remke,Eric Bouffet,Simon Bailey,Steven C. Clifford,François Doz,Marcel Kool,Christelle Dufour,Gilles Vassal,Till Milde,Olaf Witt,Katja von Hoff,Torsten Pietsch,Paul A. Northcott,Amar Gajjar,Giles W. Robinson,Laetitia Padovani,Nicolas André,Maura Massimino,Barry Pizer,Roger J. Packer,Stefan Rutkowski,Stefan M. Pfister,Michael D. Taylor,Scott L. Pomeroy +24 more
TL;DR: A consensus can serve as an outline for prioritization of certain molecular subsets of tumors to define and validate risk groups as a basis for future clinical trials.
Journal ArticleDOI
Results of a Randomized Study of Preradiation Chemotherapy Versus Radiotherapy Alone for Nonmetastatic Medulloblastoma: The International Society of Paediatric Oncology/United Kingdom Children’s Cancer Study Group PNET-3 Study
Roger E. Taylor,Clifford C. Bailey,K. Robinson,Claire Weston,David W. Ellison,James W. Ironside,H Lucraft,Richard J. Gilbertson,Diana Tait,David Walker,Barry Pizer,J. Imeson,Linda S Lashford +12 more
TL;DR: This is the first large multicenter randomized study to demonstrate improved EFS for chemotherapy compared with RT alone, and it is anticipated that this regimen could reduce ototoxicity and nephrotoxicity compared with cisplatin-containing schedules.
Journal ArticleDOI
Novel molecular subgroups for clinical classification and outcome prediction in childhood medulloblastoma: a cohort study
Edward C. Schwalbe,Edward C. Schwalbe,Janet C. Lindsey,Sirintra Nakjang,Stephen Crosier,Amanda Smith,Debbie Hicks,Gholamreza Rafiee,Rebecca M Hill,Alice Iliasova,Alice Iliasova,Thomas J Stone,Barry Pizer,Antony Michalski,Abhijit Joshi,Stephen B. Wharton,Thomas S. Jacques,Simon Bailey,Daniel Williamson,Steven C. Clifford +19 more
TL;DR: The discovery of seven novel, clinically significant subgroups of childhood medulloblastoma improves disease risk-stratification and could inform treatment decisions and provide a new foundation for future research and clinical investigations.
Journal ArticleDOI
DICER1 syndrome: clarifying the diagnosis, clinical features and management implications of a pleiotropic tumour predisposition syndrome
Ingrid Slade,Chiara Bacchelli,Helen Davies,Anne Murray,Fatemeh Abbaszadeh,Sandra Hanks,Rita Barfoot,Amos Burke,Julia C. Chisholm,Martin Hewitt,Helen Jenkinson,Derek J. King,Bruce Morland,Barry Pizer,Katrina Prescott,Anand Saggar,Lucy Side,Heidi Traunecker,Sucheta Vaidya,Paul Ward,P. Andrew Futreal,Gordan M. Vujanic,Andrew G. Nicholson,Neil J. Sebire,Clare Turnbull,John R. Priest,Kathryn Pritchard-Jones,Richard S. Houlston,Charles A. Stiller,Michael R. Stratton,Jenny Douglas,Nazneen Rahman +31 more
TL;DR: Constitutional DICER1 haploinsufficiency predisposes to a broad range of tumours, making a substantial contribution to PPB, cystic nephroma and ovarian Sertoli–Leydig tumour, but a smaller contribution to other tumours.
Journal ArticleDOI
Hyperfractionated Versus Conventional Radiotherapy Followed by Chemotherapy in Standard-Risk Medulloblastoma: Results From the Randomized Multicenter HIT-SIOP PNET 4 Trial
Birgitta Lannering,Stefan Rutkowski,François Doz,Barry Pizer,Göran Gustafsson,Aurora Navajas,Maura Massimino,Roel E. Reddingius,Martin Benesch,Christian Carrie,Roger E. Taylor,Lorenza Gandola,Thomas Björk-Eriksson,Jordi Giralt,Foppe Oldenburger,Torsten Pietsch,Dominique Figarella-Branger,Keith Robson,Marco Forni,Steven C. Clifford,M. Warmuth-Metz,Katja von Hoff,Andreas Faldum,Véronique Mosseri,Rolf D. Kortmann +24 more
TL;DR: In this paper, the authors compared the performance of standard-risk medulloblastoma patients treated by post-operative hyperfractionated or conventionally fractionated radiotherapy followed by maintenance chemotherapy.
