Mitofusin 2 in POMC Neurons Connects ER Stress with Leptin Resistance and Energy Imbalance
Marc Schneeberger,Marcelo O. Dietrich,Marcelo O. Dietrich,David Sebastián,Monica Imbernon,Carlos Castaño,Ainhoa Garcia,Yaiza Esteban,Alba Gonzalez-Franquesa,Ignacio Rodríguez,Analía Bortolozzi,Pablo M. Garcia-Roves,Ramon Gomis,Rubén Nogueiras,Tamas L. Horvath,Antonio Zorzano,Marc Claret +16 more
TL;DR: It is shown that mitochondria-ER contacts in anorexigenic pro-opiomelanocortin neurons in the hypothalamus are decreased in diet-induced obesity, establishing MFN2 in POMC neurons as an essential regulator of systemic energy balance by fine-tuning the mitochondrial-ER axis homeostasis and function.
Journal ArticleDOI
Obesity-associated exosomal miRNAs modulate glucose and lipid metabolism in mice
TL;DR: The results support a role for exosomal miRNAs in the modulation of glucose and lipid metabolism in mice and may help to uncover thus far unexplored pathological mechanisms and provide us with novel therapeutic targets.
Journal ArticleDOI
Recessive mutations in the INS gene result in neonatal diabetes through reduced insulin biosynthesis
Intza Garin,Emma L. Edghill,Ildem Akerman,Oscar Rubio-Cabezas,Itxaso Rica,Jonathan M. Locke,Miguel A. Maestro,Adnan Alshaikh,Rüveyde Bundak,Gabriel del Castillo,Asma Deeb,Dorothee Deiss,Juan M. Fernandez,Koumudi Godbole,Khalid Hussain,Michele O’Connell,Thomasz Klupa,Stanislava Kolouskova,Fauzia Mohsin,Kusiel Perlman,Zdenek Sumnik,Jose M. Rial,Estibaliz Ugarte,Thiruvengadam Vasanthi,Karen A. Johnstone,Sarah E. Flanagan,Rosa de Diego Martínez,Carlos Castaño,Ann-Marie Patch,Eduardo Fernandez-Rebollo,Klemens Raile,Noel G. Morgan,Lorna W. Harries,Luis Castaño,Sian Ellard,Jorge Ferrer,Guiomar Perez de Nanclares,Andrew T. Hattersley +37 more
TL;DR: Ten recessively inherited mutations in the INS gene that encodes preproinsulin provide a genetic demonstration of the essential role of multiple sequence elements that regulate the biosynthesis of insulin in man.
Journal ArticleDOI
Deletion of miRNA processing enzyme Dicer in POMC-expressing cells leads to pituitary dysfunction, neurodegeneration and development of obesity
Marc Schneeberger,Jordi Altirriba,Ainhoa Garcia,Yaiza Esteban,Carlos Castaño,Montserrat Garcia-Lavandeira,Clara V. Alvarez,Ramon Gomis,Marc Claret +8 more
TL;DR: The expression of Dicer, an essential endoribonuclease for miRNA maturation, is modulated by nutrient availability and excess in the hypothalamus, which highlights a key role for miRNAs in POMC neuron survival and the consequent development of neurodegenerative obesity.
Journal ArticleDOI
Exosomes and diabetes
TL;DR: The molecular dissection of the pathways initiated by exosomes under situations of metabolic stress could help to gain a deeper knowledge of the pathophysiology of diabetes and its associated metabolic diseases.
