Aberrant Cleavage of TDP-43 Enhances Aggregation and Cellular Toxicity
Yong Jie Zhang,Ya Fei Xu,Casey Cook,Tania F. Gendron,Paul S. Roettges,Christopher D. Link,Wen Lang Lin,Jimei Tong,Monica Castanedes-Casey,Peter E.A. Ash,Jennifer Gass,Vijayaraghavan Rangachari,Vijayaraghavan Rangachari,Emanuele Buratti,Francisco E. Baralle,Todd E. Golde,Dennis W. Dickson,Leonard Petrucelli +17 more
TL;DR: It is reported that the ectopic expression of a ≈25-kDa TDP-43 fragment corresponding to the C-terminal truncation product of caspase-cleaved T DP-43 leads to the formation of toxic, insoluble, and ubiquitin- and phospho-positive cytoplasmic inclusions within cells.
Journal ArticleDOI
Neuronal activity enhances tau propagation and tau pathology in vivo
Jessica W. Wu,S. Abid Hussaini,Isle M Bastille,Gustavo A. Rodriguez,Ana Mrejeru,Kelly Rilett,David W. Sanders,Casey Cook,Hongjun Fu,Rick A C M Boonen,Mathieu Herman,Eden Nahmani,Sheina Emrani,Y Helen Figueroa,Marc I. Diamond,Catherine L. Clelland,Selina Wray,Karen Duff +17 more
TL;DR: It is shown that physiological tau released from donor cells can transfer to recipient cells via the medium, suggesting that at least one mechanism by which tau can transfer is via the extracellular space.
Journal ArticleDOI
An autoradiographic evaluation of AV-1451 Tau PET in dementia.
Val J. Lowe,Geoffry L. Curran,Ping Fang,Amanda M. Liesinger,Keith A. Josephs,Joseph E. Parisi,Kejal Kantarci,Bradley F. Boeve,Mukesh K. Pandey,Tyler J. Bruinsma,David S. Knopman,David T.W. Jones,Leonard Petrucelli,Casey Cook,Neill R. Graff-Radford,Dennis W. Dickson,Ronald C. Petersen,Clifford R. Jack,Melissa E. Murray +18 more
TL;DR: Reduced AV-1451 binding in neuritic pathology compared to neurofibrillary tangles suggests that the maturity of tau pathology may affect AV- fourteen51 binding and suggests complexity in AV- 1451 binding.
Journal ArticleDOI
Posttranslational Modifications Mediate the Structural Diversity of Tauopathy Strains.
Tamta Arakhamia,Christina E. Lee,Yari Carlomagno,Duc M. Duong,Sean R. Kundinger,Kevin Wang,Dewight Williams,Michael DeTure,Dennis W. Dickson,Casey Cook,Nicholas T. Seyfried,Leonard Petrucelli,Anthony W. P. Fitzpatrick +12 more
TL;DR: Cryo-electron microscopy is used to determine the structures of tau filaments from corticobasal degeneration (CBD) human brain tissue and proposes a structure-based model in which cross-talk between PTMs influences tau filament structure, contributing to the structural diversity of tauopathy strains.
Journal ArticleDOI
Tau Protein Disrupts Nucleocytoplasmic Transport in Alzheimer's Disease
Bahareh Eftekharzadeh,J. Gavin Daigle,Larisa E. Kapinos,Alyssa N. Coyne,Julia Schiantarelli,Yari Carlomagno,Casey Cook,Sean J. Miller,Simon Dujardin,Ana S. Amaral,Jonathan C. Grima,Rachel E. Bennett,Katharina Tepper,Michael DeTure,Charles R. Vanderburg,Bianca T. Corjuc,Sarah L. DeVos,Jose J. Gonzalez,Jeannie Chew,Svetlana Vidensky,Fred H. Gage,Jerome Mertens,Juan C. Troncoso,Eckhard Mandelkow,Xavier Salvatella,Roderick Y. H. Lim,Leonard Petrucelli,Susanne Wegmann,Jeffrey D. Rothstein,Bradley T. Hyman +29 more
TL;DR: It is shown that tau can directly interact with nucleoporins of the nuclear pore complex (NPC) and affect their structural and functional integrity and raises the possibility that NPC dysfunction contributes to tau-induced neurotoxicity in AD and tauopathies.
