Journal ArticleDOI
SIRT3 regulates mitochondrial fatty-acid oxidation by reversible enzyme deacetylation
Matthew D. Hirschey,Tadahiro Shimazu,Tadahiro Shimazu,Eric S. Goetzman,Enxuan Jing,Bjoern Schwer,Bjoern Schwer,Bjoern Schwer,David B. Lombard,Carrie A. Grueter,Charles A. Harris,Sudha B. Biddinger,Olga Ilkayeva,Robert Stevens,Yu Li,Asish K. Saha,Neil B. Ruderman,James R. Bain,Christopher B. Newgard,Robert V. Farese,Frederick W. Alt,C. Ronald Kahn,Eric Verdin,Eric Verdin +23 more
TL;DR: It is demonstrated that SIRT3 modulates mitochondrial intermediary metabolism and fatty-acid use during fasting and acetylation is identified as a novel regulatory mechanism for mitochondrial fatty- acid oxidation.
Journal ArticleDOI
Thematic review series: glycerolipids. DGAT enzymes and triacylglycerol biosynthesis.
TL;DR: The genes encoding two DGAT enzymes, DGAT1 and DGAT2, were identified in the past decade, and the use of molecular tools, including mice deficient in either enzyme, has shed light on their functions.
Journal ArticleDOI
Efficient hepatitis C virus particle formation requires diacylglycerol acyltransferase-1.
Eva Herker,Charles A. Harris,Céline Hernandez,Arnaud Carpentier,Katrin Kaehlcke,Arielle R. Rosenberg,Robert V. Farese,Melanie Ott +7 more
TL;DR: This work identifies the triglyceride-synthesizing enzyme diacylglycerol acyltransferase-1 (DGAT1) as a key host factor for HCV infection and implicates DGAT1 as a new target for antiviral therapy.
Journal ArticleDOI
BH3-only Bcl-2 family members are coordinately regulated by the JNK pathway and require Bax to induce apoptosis in neurons.
TL;DR: It is shown that the BH3-only Bcl-2 family members, Dp5/Hrk and Bim, are induced upstream of the Bax checkpoint in neuronal apoptosis in a manner that shows significant dependence on JNK signaling.
Journal ArticleDOI
DGAT enzymes are required for triacylglycerol synthesis and lipid droplets in adipocytes
Charles A. Harris,Joel T. Haas,Ryan S. Streeper,S. Stone,Manju Kumari,Kui Yang,Xianlin Han,Nicholas Brownell,Richard W. Gross,Rudolf Zechner,Robert V. Farese +10 more
TL;DR: It is shown that DGAT1 and DGAT2 account for the vast majority of TG synthesis in mice, and DG AT function is required for LDs in adipocytes, but not in all cell types.
