Journal ArticleDOI
Restoration of metabolic health by decreased consumption of branched-chain amino acids.
Nicole E. Cummings,Elizabeth Williams,Ildiko Kasza,Elizabeth N. Konon,Michael D. Schaid,Brian A. Schmidt,Chetan Poudel,Dawn S. Sherman,Deyang Yu,Sebastian I. Arriola Apelo,Sara E. Cottrell,Gabriella Geiger,Macy E. Barnes,Jaclyn A. Wisinski,Rachel J. Fenske,Kristina A. Matkowskyj,Michelle E. Kimple,Caroline M. Alexander,Matthew J. Merrins,Dudley W. Lamming +19 more
TL;DR: It is shown that a reduced BCAA diet promotes rapid fat mass loss without calorie restriction in obese mice, and it is suggested that dietary protein quality (i.e. the precise macronutrient composition of dietary protein) may impact the effectiveness of weight loss diets.
Journal ArticleDOI
Pyruvate Kinase Controls Signal Strength in the Insulin Secretory Pathway
Sophie L. Lewandowski,Rebecca L. Cardone,Hannah R. Foster,Thuong Ho,Evgeniy Potapenko,Chetan Poudel,Halena R. VanDeusen,Sophia M. Sdao,Tiago C. Alves,Xiaojian Zhao,Megan E. Capozzi,Arnaldo H. de Souza,Ishrat Jahan,Craig J. Thomas,Craig S. Nunemaker,Dawn Belt Davis,Jonathan E. Campbell,Richard G. Kibbey,Matthew J. Merrins +18 more
TL;DR: It is shown that pyruvate kinase (PK) underlies β cell sensing of both glycolytic and mitochondrial fuels and suggests a potential therapeutic route for diabetes based on PK activation that would not be predicted by the current consensus single-state model of β cell function.
Journal ArticleDOI
Pancreatic β-Cells From Mice Offset Age-Associated Mitochondrial Deficiency With Reduced KATP Channel Activity.
Trillian Gregg,Chetan Poudel,Brian A. Schmidt,Rashpal S. Dhillon,Sophia M. Sdao,Nathan A. Truchan,Emma L. Baar,Luis A. Fernandez,John M. Denu,Kevin W. Eliceiri,Jeremy D. Rogers,Michelle E. Kimple,Dudley W. Lamming,Matthew J. Merrins +13 more
TL;DR: It is demonstrated how mice but not humans are able to successfully compensate for age-associated metabolic dysfunction by adjusting β-cell glucose sensitivity and highlight an essential mechanism for ensuring the maintenance of insulin secretion.
Posted ContentDOI
Observation of an α-synuclein liquid droplet state and its maturation into Lewy body-like assemblies
Maarten C. Hardenberg,Tessa Sinnige,Sam Casford,Samuel T. Dada,Chetan Poudel,Lizzy Robinson,Monika Fuxreiter,Clemens F Kaminksi,Gabriele S. Kaminski Schierle,Ellen A. A. Nollen,Christopher M. Dobson,Michele Vendruscolo +11 more
TL;DR: It is shown that α-synuclein undergoes liquid-liquid phase separation by forming a liquid droplet state, which converts into an amyloid-rich hydrogel, and that this maturation process towards the amyloids state is delayed in the presence of model synaptic vesicles in vitro.
Journal ArticleDOI
A Highly Porous Metal-Organic Framework System to Deliver Payloads for Gene Knockdown
Michelle H. Teplensky,Marcus Fantham,Chetan Poudel,Colin Hockings,Meng Lu,Alina Guna,Marta Aragones-Anglada,Peyman Z. Moghadam,Peyman Z. Moghadam,Peng Li,Omar K. Farha,Sandra Bernaldo de Quirós Fernández,Frances M. Richards,Duncan I. Jodrell,Gabriele Kaminski Schierle,Clemens F. Kaminski,David Fairen-Jimenez +16 more
TL;DR: The use of a metal-organic framework (MOF) to load, protect, and deliver small interfering ribonucleic acids (siRNAs) is reported and the protection of MOF-internalized siRNA from enzymatic degradation is confirmed.
