Journal ArticleDOI

An orally available Mpro inhibitor is effective against wild-type SARS-CoV-2 and variants including Omicron

TL;DR: In this article , a series of potent α-ketoamide-containing Mpro inhibitors were obtained using the Ugi four-component reaction and the prioritized compound, Y180, showed an IC50 of 8.1 nM against SARS-CoV-2 Mpro and had oral bioavailability of 92.9%, 31.9% and 85.7% in mice, rats and dogs, respectively.
Journal ArticleDOI

Discovery of 5-(5-fluoro-1H-pyrrolo[2,3-b]pyridin-3-yl)pyrazin-2(1H)-one derivatives as new potent PB2 inhibitors

TL;DR: The discovery of a new series of PB2 inhibitors containing the skeleton 5-fluoro-1H-pyrrolo[2,3-b]pyridin-3-yl)pyrazin-2(1H)-one provides a promising lead compound for subsequent anti-influenza drug discovery targeting PB2.
Journal ArticleDOI

Non-classical ferroptosis inhibition by a small molecule targeting PHB2

TL;DR: Li et al. as mentioned in this paper reported a new ferroptosis inhibitor, termed YL-939, which is neither an antioxidant nor an iron-chelator, and revealed a new regulation mechanism of FerroPTosis.
Journal ArticleDOI

Discovery of 3,4-Dihydrobenzo[f][1,4]oxazepin-5(2H)-one Derivatives as a New Class of Selective TNIK Inhibitors and Evaluation of Their Anti-Colorectal Cancer Effects.

TL;DR: Structural-activity relationship analyses led to the identification of a number of potent TNIK inhibitors with compound 21k being the most active one and displaying excellent selectivity for TNIK against 406 other kinases.
