Journal ArticleDOI

Venetoclax with azacitidine disrupts energy metabolism and targets leukemia stem cells in patients with acute myeloid leukemia.

TL;DR: This report shows that treatment of older patients with AML with the B cell lymphoma 2 (BCL-2) inhibitor venetoclax in combination with azacitidine results in deep and durable remissions and is superior to conventional treatments.
Journal ArticleDOI

Chronic myeloid leukemia stem cells possess multiple unique features of resistance to BCR-ABL targeted therapies.

TL;DR: It is shown that the lesser effect of imatinib mesylate on the 3-week output of cells produced in vitro from lin−CD34+CD38− CML (stem) cells compared with cultures initiated with the CD38+ subset of lin− CD34+ cells is markedly enhanced when conditions of reduced growth factor stimulation are used.
Journal ArticleDOI

Inhibition of Amino Acid Metabolism Selectively Targets Human Leukemia Stem Cells.

TL;DR: It is demonstrated that amino acid uptake, steady-state levels, and catabolism are all elevated in the leukemia stem cell (LSC) population, indicating that clinically relevant eradication of LSCs can be achieved with drugs that target LSC metabolic vulnerabilities.
Journal ArticleDOI

Prospective isolation and molecular characterization of hematopoietic stem cells with durable self-renewal potential.

TL;DR: It is shown that cells capable of repopulating irradiated congenic hosts for 4 months and producing clones of cells that can be serially transplanted are selectively and highly enriched in the CD150(+) subset of the EPCR(+)CD48(-)CD45(+) fraction of mouse fetal liver and adult bone marrow cells.
