Journal ArticleDOI
Endoplasmic reticulum stress signalling and the pathogenesis of non-alcoholic fatty liver disease
Cynthia Lebeaupin,Déborah Vallée,Younis Hazari,Claudio Hetz,Eric Chevet,Béatrice Bailly-Maitre +5 more
TL;DR: Experimental data suggest that the unfolded protein response influences hepatic tumour development, aggressiveness and response to treatment, offering novel therapeutic avenues, and possible points of intervention are discussed.
Journal ArticleDOI
ER stress induces NLRP3 inflammasome activation and hepatocyte death.
Cynthia Lebeaupin,Cynthia Lebeaupin,E. Proics,E. Proics,C.-H.D. de Bieville,C.-H.D. de Bieville,Déborah Rousseau,Déborah Rousseau,S. Bonnafous,Stéphanie Patouraux,G Adam,G Adam,Vanessa Lavallard,Vanessa Lavallard,Carole Rovère,O. Le Thuc,Marie-Christine Saint-Paul,Rodolphe Anty,Anne-Sophie Schneck,Antonio Iannelli,Jean Gugenheim,Albert Tran,Philippe Gual,Philippe Gual,Beatrice Bailly-Maitre,Beatrice Bailly-Maitre +25 more
TL;DR: It is demonstrated here that ER stress leads to hepatic NLRP3 inflammasome pyroptotic death, thus contributing as a novel mechanism of inflammation-mediated liver injury in chronic liver diseases.
Journal ArticleDOI
Interactome Screening Identifies the ER Luminal Chaperone Hsp47 as a Regulator of the Unfolded Protein Response Transducer IRE1α
Denisse Sepulveda,Diego Rojas-Rivera,Diego A. Rodriguez,Diego A. Rodriguez,Jody Groenendyk,Andres Köhler,Cynthia Lebeaupin,Shinya Ito,Hery Urra,Amado Carreras-Sureda,Younis Hazari,Mireille Vasseur-Cognet,Maruf M.U. Ali,Eric Chevet,G Campos,Patricio Godoy,Tomas Vaisar,Béatrice Bailly-Maitre,Kazuhiro Nagata,Marek Michalak,Jimena Sierralta,Claudio Hetz +21 more
TL;DR: It is concluded that Hsp47 adjusts IRE1α signaling by fine-tuning the threshold to engage an adaptive UPR.
Journal ArticleDOI
CD44 is a key player in non-alcoholic steatohepatitis
Stéphanie Patouraux,Déborah Rousseau,Stéphanie Bonnafous,Cynthia Lebeaupin,Carmelo Luci,Clémence M. Canivet,Anne-Sophie Schneck,Adeline Bertola,Marie-Christine Saint-Paul,Antonio Iannelli,Jean Gugenheim,Rodolphe Anty,Albert Tran,Béatrice Bailly-Maitre,Philippe Gual +14 more
TL;DR: Human and experimental data suggest that CD44, a cellular protein mainly expressed in immune cells, is a marker and key player of hepatic inflammation and its targeting partially corrects NASH, making it a potential therapeutic strategy.
Journal ArticleDOI
Low-Protein Diet Induces IRE1α-Dependent Anticancer Immunosurveillance
Camila Rubio-Patiño,Jozef P. Bossowski,Gian Marco De Donatis,Laura Mondragón,Elodie Villa,Lazaro Emilio Aira,Johanna Chiche,Rana Mhaidly,Cynthia Lebeaupin,Sandrine Marchetti,Konstantinos Voutetakis,Aristotelis Chatziioannou,Florence Castelli,Patricia Lamourette,Emeline Chu-Van,François Fenaille,Tony Avril,Thierry Passeron,John B. Patterson,Els Verhoeyen,Béatrice Bailly-Maitre,Eric Chevet,Jean-Ehrland Ricci +22 more
TL;DR: It is demonstrated that feeding mice a low-protein (Low PROT) isocaloric diet but not aLow CHO diet reduced tumor growth in three independent mouse cancer models, suggesting that a Low PROT diet induces an IRE1α-dependent UPR in cancer cells, enhancing a CD8-mediated T cell response against tumors.
