Heavy metal contamination of soil and water causing toxicity/stress has become one important constraint to crop productivity and quality. This situation has further worsened by the increasing population growth and inherent food demand. It have been reported in several studies that counterbalancing toxicity, due to heavy metal requires complex mechanisms at molecular, biochemical, physiological, cellular, tissue and whole plant level, which might manifest in terms of improved crop productivity. Recent advances in various disciplines of biological sciences such as metabolomics, transcriptomics, proteomics etc. have assisted in the characterization of metabolites, transcription factors, stress-inducible proteins involved in heavy metal tolerance, which in turn can be utilized for generating heavy metal tolerant crops. This review summarizes various tolerance strategies of plants under heavy metal toxicity, covering the role of metabolites (metabolomics), trace elements (ionomics), transcription factors (transcriptomics), various stress-inducible proteins (proteomics) as well as the role of plant hormones. We also provide a glance at strategies adopted by metal accumulating plants also known as “metallophytes”.

Plants are the target of a wide range of pollutants that vary in concentration, speciation, and toxicity. Such pollutants mainly enter the plant system through the soil (Arshad et al. 2008) or via the atmosphere (Uzu et al. 2010). Among common pollutants that affect plants, lead is among the most toxic and frequently encountered (Cecchi et al. 2008; Grover et al. 2010; Shahid et al. 2011). Lead continues to be used widely in many industrial processes and occurs as a contaminant in all environmental compartments (soils, water, the atmosphere, and living organisms). The prominence of environmental lead contamination results both from its persistence (Islam et al. 2008; Andra et al. 2009; Punamiya et al. 2010) and from its present and past numerous sources. These sources have included smelting, combustion of leaded gasoline, or applications of lead-contaminated media (sewage sludge and fertilizers) to land (Piotrowska et al. 2009; Gupta et al. 2009; Sammut et al. 2010; Grover et al. 2010). In 2009, production of recoverable lead from mining operations was 1690, 516, and 400 thousand metric tons by China, Australia, and the USA, respectively (USGS 2009).

Lead Uptake, Toxicity, and Detoxification in Plants

Wild sweet cherry (Prunus avium) trees are abundant in the northern part of Turkey, including the Coruh Valley. We analyzed 18 wild sweet cherry genotypes collected from diverse environments in the upper Coruh Valley in Turkey to determine genetic variation, using 10 SSR primers. These SSR primers generated 46 alleles; the number of alleles per primer ranged from 3 to 7, with a mean of 4.6. The primer PS12A02 gave the highest number of polymorphic bands (N = 7), while CPSCT010, UDAp-401 and UDAp-404 gave the lowest number (N = 3). Seven groups were separated in the dendrogram, although most of the genotypes did not cluster according to phenological and morphological traits. This level of genetic diversity in these wild sweet cherry genotypes is very high and therefore these trees would be useful as breeders for crosses between cultivated sweet cherry and wild genotypes.

Genetic diversity in wild sweet cherries (Prunus avium) in Turkey revealed by SSR markers.

Assessment of DNA damages stemming from toxic chemicals is an important issue in terms of genotoxicology. In this study, maize (Zea mays L.) seedlings were used for screening the genotoxic effects of cobalt (Co) and nickel (Ni) treatments at various concentrations (5 mM, 10 mM, 20 mM and 40 mM). For this purpose, randomly amplified polymorphic DNA (RAPD) technique was applied to genomic DNA extracted from metal-exposed and unexposed plant materials. Besides, changes in total protein contents were screened by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis. For RAPD analysis, 16 RAPD primers were found to produce unique polymorphic band profiles on different concentrations of Co-/Ni-treated maize seedlings. Increased polymorphism resulting from the appearance of new bands or disappearance of normal bands was observed with increasing concentration of Co and Ni treatments. Genomic template stability, a qualitative measurement of changes in RAPD patterns of genomic DNA, decreased with increasing metal concentration. In SDS-PAGE analysis, it was observed that the total soluble protein content decreased by Co treatment, while it increased by Ni treatment. The results obtained from this study revealed that RAPD profiles and total soluble protein levels can be applied to detect genotoxicity, and these analyses can offer useful biomarker assays for the evaluation of genotoxic effects on Co- and Ni-polluted plants.

Molecular determination of genotoxic effects of cobalt and nickel on maize (Zea mays L.) by RAPD and protein analyses.

This study was carried out to determine genetic relationships among 23 apricot cultivars from Turkey by using randomly amplified polymorphic DNA (RAPD) technology. Among the 40 decamer random primers were used for PCR reactions, 12 primers showed reproducible polymorphic patterns. These primers produced 121 bands, out of which 118 were polymorphic. UPGMA (Unweighted Pair Group Method using Arithmetic average) derived dendrogram, grouped the cultivars into one cluster and one outlier. The main cluster further divided into three subclusters. The highest dissimilarities found between Hacihaliloglu and Kabaasi cultivars (0.108). The variation detected among the closely related genotypes indicates the efficiency of RAPD markers over the morphological and isozyme markers for the identification and construction of genetic linkage maps.
