Activation by IKKα of a Second, Evolutionary Conserved, NF-κB Signaling Pathway
Uwe Senftleben,Yixue Cao,Gutian Xiao,Florian R. Greten,Gertraud Krähn,Giuseppina Bonizzi,Yi Chen,Yinling Hu,Abraham Fong,Shao Cong Sun,Michael Karin +10 more
TL;DR: It is shown that IKKα is required for B cell maturation, formation of secondary lymphoid organs, increased expression of certain NF-κB target genes, and processing of the NF-σκB2 (p100) precursor.
Journal ArticleDOI
NF-κB-Inducing Kinase Regulates the Processing of NF-κB2 p100
TL;DR: The data suggest that NIK is a specific kinase regulating p100 processing and explain why the aly and nf(kappa)b2 knockout mice exhibit similar immune deficiencies.
Journal ArticleDOI
Retroviral oncoprotein Tax induces processing of NF‐κB2/p100 in T cells: evidence for the involvement of IKKα
Gutian Xiao,Mary Ellen Cvijic,Abraham Fong,Edward W. Harhaj,Mark Uhlik,Michael Waterfield,Shao Cong Sun +6 more
TL;DR: It is shown that in T cells infected with the human T‐cell leukemia virus (HTLV), IKKα is targeted to a novel signaling pathway that mediates processing of the nfκb2 precursor protein p100, resulting in active production of the NF‐κB subunit, p52, which is mediated by the HTLV‐encoded oncoprotein Tax.
Journal ArticleDOI
NF-κB-inducing Kinase and IκB Kinase Participate in Human T-cell Leukemia Virus I Tax-mediated NF-κB Activation
Mark Uhlik,Li Feng Good,Gutian Xiao,Edward W. Harhaj,Ebrahim Zandi,Michael Karin,Shao Cong Sun +6 more
TL;DR: It is demonstrated that Tax triggers activation of cellular protein kinases, IκB kinase α (IKKα) and IKKβ, which phosphorylate the NF-κB inhibitory protein IKKBα, resulting in its degradation and NF-σκB activation.
Journal ArticleDOI
Somatic mutagenesis studies of NF-κB signaling in human T cells: Evidence for an essential role of IKKγ, in NP-κB activation by T-cell costimulatory signals and HTLV-I Tax protein
Edward W. Harhaj,Li Feng Good,Gutian Xiao,Mark Uhlik,Mary Ellen Cvijic,Isis Rivera-Walsh,Shao Cong Sun +6 more
TL;DR: Genetic evidence is provided for the requirement of IKKγ in NF-κB signaling triggered by both T-cell costimulatory signals and HTLV-I Tax protein.
