Journal ArticleDOI
Deciphering protein kinase specificity through large-scale analysis of yeast phosphorylation site motifs
Janine Mok,Philip M. Kim,Philip M. Kim,Hugo Y. K. Lam,Stacy Piccirillo,Xiuqiong Zhou,Grace R. Jeschke,Douglas L. Sheridan,Douglas L. Sheridan,Sirlester A. Parker,Ved Desai,Miri Jwa,Elisabetta Cameroni,Hengyao Niu,Matthew C. Good,Attila Remenyi,Attila Remenyi,Jia Lin Nianhan Ma,Yi-Jun Sheu,Holly E. Sassi,Richelle Sopko,Clarence S.M. Chan,Claudio De Virgilio,Nancy M. Hollingsworth,Wendell A. Lim,David F. Stern,Bruce Stillman,Brenda J. Andrews,Mark Gerstein,Michael Snyder,Michael Snyder,Benjamin E. Turk +31 more
TL;DR: A rapid peptide screening approach was used to determine consensus phosphorylation site motifs targeted by 61 of the 122 kinases in Saccharomyces cerevisiae, and previously unappreciated rules for determining specificity within the kinase family were uncovered.
Journal ArticleDOI
Mechanism of the ATP-dependent DNA end-resection machinery from Saccharomyces cerevisiae
Hengyao Niu,Woo Hyun Chung,Zhu Zhu,Youngho Kwon,Weixing Zhao,Peter Chi,Rohit Prakash,Changhyun Seong,Dongqing Liu,Lucy Lu,Grzegorz Ira,Patrick Sung +11 more
TL;DR: It is shown that DNA strand separation during end resection is mediated by the Sgs1 helicase function, in a manner that is enhanced by Top3–Rmi1 and MRX, and a multifaceted role of RPA is unveiled, in the sequestration of ssDNA generated by DNA unwinding, enhancement of 5′ strand incision, and protection of the 3′ strand.
Journal ArticleDOI
Pif1 helicase and Polδ promote recombination-coupled DNA synthesis via bubble migration
Marenda A. Wilson,Youngho Kwon,Yuanyuan Xu,Woo-Hyun Chung,Woo-Hyun Chung,Woo-Hyun Chung,Peter Chi,Peter Chi,Hengyao Niu,Ryan Mayle,Xuefeng Chen,Anna Malkova,Anna Malkova,Patrick Sung,Grzegorz Ira +14 more
TL;DR: Purified Pif1 protein strongly stimulates Polδ-mediated DNA synthesis from a D-loop made by the Rad51 recombinase, uncovering a novel function of P if1 and providing insights into the mechanism of HR.
Journal ArticleDOI
Partner choice during meiosis is regulated by Hop1-promoted dimerization of Mek1
Hengyao Niu,Lihong Wan,Bridget L. Baumgartner,Dana Schaefer,Josef Loidl,Nancy M. Hollingsworth +5 more
TL;DR: A model in which Hop1 phosphorylation in response to DSBs triggers dimerization of Mek1 via the Hop1 C domain, thereby enabling Mek1 to phosphorylate target proteins that prevent repair of D SBs by sister chromatids is suggested.
Journal ArticleDOI
Concentration-Dependent Exchange of Replication Protein a on Single-Stranded DNA Revealed by Single-Molecule Imaging
Bryan Gibb,Ling Feng Ye,Stephanie C. Gergoudis,Youngho Kwon,Hengyao Niu,Patrick Sung,Eric C. Greene +6 more
TL;DR: The results reveal an unanticipated exchange between bound and free RPA suggesting a binding mechanism that can confer exceptionally slow off rates, yet also enables rapid displacement through a direct exchange mechanism that is reliant upon the presence of free ssDNA-binding proteins in solution.
