A Distinct Small RNA Pathway Silences Selfish Genetic Elements in the Germline
Vasily V. Vagin,Alla A. Sigova,Chengjian Li,Hervé Seitz,Vladimir A. Gvozdev,Phillip D. Zamore +5 more
TL;DR: The data suggest that rasiRNAs protect the fly germline through a silencing mechanism distinct from both the miRNA and RNA interference pathways.
Journal ArticleDOI
Endogenous siRNAs Derived from Transposons and mRNAs in Drosophila Somatic Cells
Megha Ghildiyal,Hervé Seitz,Michael D. Horwich,Chengjian Li,Tingting Du,Soohyun Lee,Jia Xu,Ellen L. W. Kittler,Maria L. Zapp,Zhiping Weng,Phillip D. Zamore +10 more
TL;DR: It is proposed that endo-siRNAs generated by the fly RNAi pathway silence selfish genetic elements in the soma, much as Piwi-interacting RNAs do in the germ line.
Journal ArticleDOI
Collapse of Germline piRNAs in the Absence of Argonaute3 Reveals Somatic piRNAs in Flies
Chengjian Li,Vasily V. Vagin,Soohyun Lee,Soohyun Lee,Jia Xu,Jia Xu,Shengmei Ma,Hualin Xi,Hualin Xi,Hervé Seitz,Michael D. Horwich,Monika Syrzycka,Barry M. Honda,Ellen L. W. Kittler,Maria L. Zapp,Carla Andrea Klattenhoff,Nadine Schulz,William E. Theurkauf,Zhiping Weng,Phillip D. Zamore +19 more
TL;DR: Strong loss-of-function mutations in ago3 are described and it is found that Ago3 acts to amplify piRNA pools and to enforce on them an antisense bias, increasing the number of piRNAs that can act to silence transposons.
Journal ArticleDOI
A Large Imprinted microRNA Gene Cluster at the Mouse Dlk1-Gtl2 Domain
Hervé Seitz,Hélène Royo,Marie-Line Bortolin,Shau-Ping Lin,Anne C. Ferguson-Smith,Jérôme Cavaillé +5 more
TL;DR: It is shown that the miRNA genes are only expressed from the maternally inherited chromosome and that their imprinted expression is regulated by an intergenic germline-derived differentially methylated region (IG-DMR) located approximately 200 kb upstream from the mi RNA cluster.
Journal ArticleDOI
Asymmetric regulation of imprinting on the maternal and paternal chromosomes at the Dlk1-Gtl2 imprinted cluster on mouse chromosome 12
Shau-Ping Lin,Neil A. Youngson,Shuji Takada,Shuji Takada,Hervé Seitz,Wolf Reik,Martina Paulsen,Martina Paulsen,Jérôme Cavaillé,Anne C. Ferguson-Smith +9 more
TL;DR: It is shown that deletion of the IG-DMR from the maternally inherited chromosome causes bidirectional loss of imprinting of all genes in the 1-Mb cluster and indicates that the two parental chromosomes control allele-specific gene expression differently.
