Journal ArticleDOI

The PI3K pathway in human disease

TL;DR: A perspective on the roles of class I PI3Ks in the regulation of cellular metabolism and in immune system functions is provided, two topics closely intertwined with cancer biology.
Journal ArticleDOI

mTOR kinase inhibitors promote antibody class switching via mTORC2 inhibition.

TL;DR: It is found that a new class of ATP-competitive mTOR kinase inhibitors (TOR-KIs) can augment antibody class switching at concentrations that partially inhibit mTOR activity, and opposing roles for mTORC1 and m TORC2 in B-cell differentiation are identified and mechanisms of mTOR signaling in B cells are uncovers.
Journal ArticleDOI

The mTORC1/4E-BP/eIF4E Axis Promotes Antibody Class Switching in B Lymphocytes.

TL;DR: A previously undescribed role for mTORC1 and the eif4E-binding proteins/eIF4E axis in activation-induced cytidine deaminase protein expression and Ab class switching in mouse B cells is uncovered, suggesting that cap-dependent translation regulates key steps in B cell differentiation.
Journal ArticleDOI

The Selective Phosphoinoside-3-Kinase p110δ Inhibitor IPI-3063 Potently Suppresses B Cell Survival, Proliferation, and Differentiation

TL;DR: Findings show that p110δ is the dominant isoform mediating B cell responses and establish that IPI-3063 is a highly potent molecule useful for studying p 110δ function in immune cells.
