Genetic regulation of human aortic smooth muscle cell gene expression and splicing predict causal coronary artery disease genes
Redouane Aherrahrou,Dillon Lue,Roland N. Perry,Yonathan Tamrat Aberra,Mohammad Daud Khan,Joon Yuhl Soh,Tiit Örd,Prosanta Singha,Huda Gilani,Ernest Diez Benavente,Doris Wong,James B. Hinkle,Lijiang Ma,Gloria M. Sheynkman,Hester M. den Ruijter,Clint L. Miller,Johan Björkegren,Minna U. Kaikkonen,Mete Civelek +18 more
TL;DR: Measurements of gene expression in SMCs isolated from the ascending aortas of 151 ethnically diverse heart transplant donors in quiescent or proliferative conditions and the association of their expression and splicing with SNP markers provide evidence for the molecular mechanisms of genetic susceptibility to CAD in distinct SMC phenotypes.
Journal ArticleDOI
The angiopoietin receptor Tie2 is atheroprotective in arterial endothelium
Andrey Anisimov,Shentong Fang,Karthik Amudhala Hemanthakumar,Tiit Örd,Kristof Van Avondt,Raphael Chevre,Anu Toropainen,Prosanta Singha,Huda Gilani,Su Duy Nguyen,Sinem Yeles Karaman,Emilia A. Korhonen,Ralf H. Adams,Hellmut G. Augustin,Katariina Öörni,Oliver Soehnlein,Minna U. Kaikkonen,Kari Alitalo +17 more
TL;DR: This paper showed that deletion of Tie2 or both Tie2 and Tie1 in the arterial endothelium promotes atherosclerosis by increasing Foxo1 nuclear localization, endothelial adhesion molecule expression and accumulation of immune cells.
Journal ArticleDOI
Genetic Regulation of SMC Gene Expression and Splicing Predict Causal CAD Genes
Redouane Aherrahrou,Dillon Lue,Robert N. Perry,Yonathan Tamrat Aberra,Mohammad Daud Khan,Joon Yuhl Soh,Tiit Örd,Prosanta Singha,Qianyi Yang,Huda Gilani,Ernest Diez Benavente,Doris Wong,James B. Hinkle,Lijiang Ma,Gloria M. Sheynkman,Hester M. den Ruijter,Clint L. Miller,Johan Björkegren,Minna U. Kaikkonen,Mete Civelek +19 more
TL;DR: In this article , the authors measured gene expression in vascular smooth muscle cells (SMCs) isolated from the ascending aortas of 151 heart transplant donors of various genetic ancestries in quiescent or proliferative conditions and calculated the association of their expression and splicing with 6.3 million imputed single-nucleotide polymorphism markers across the genome.
