Journal ArticleDOI
A rapid and scalable density gradient purification method for Plasmodium sporozoites
Mark Kennedy,Matthew Fishbaugher,Ashley M. Vaughan,Rapatbhorn Patrapuvich,Rachasak Boonhok,Narathatai Yimamnuaychok,Nastaran Rezakhani,Peter G. Metzger,Marisa Ponpuak,Jetsumon Sattabongkot,Stefan H. I. Kappe,Stefan H. I. Kappe,Jen C. C. Hume,Scott E. Lindner +13 more
TL;DR: A discontinuous density gradient purification method for Plasmodium sporozoites is described that maintains parasite infectivity in vitro and in vivo and greatly reduces mosquito and microbial contaminants.
Journal ArticleDOI
Development of potent and selective Plasmodium falciparum calcium-dependent protein kinase 4 (PfCDPK4) inhibitors that block the transmission of malaria to mosquitoes.
Rama Subba Rao Vidadala,Kayode K. Ojo,Steven M. Johnson,Zhongsheng Zhang,Stephen E. Leonard,Arinjay K. Mitra,Ryan Choi,Molly C. Reid,Katelyn R. Keyloun,Anna M. W. Fox,Mark Kennedy,Tiffany Silver-Brace,Jen C. C. Hume,Stefan H. I. Kappe,Christophe L. M. J. Verlinde,Erkang Fan,Ethan A. Merritt,Wesley C. Van Voorhis,Dustin J. Maly +18 more
TL;DR: It is demonstrated that pyrazolopyrimidine- and imidazopyrazine-based inhibitors that are potent inhibitors of the in vitro activity of PfCDPK4 are able to block Plasmodium falciparum exflagellation with no observable toxicity to human cells.
Journal ArticleDOI
A transgenic Plasmodium falciparum NF54 strain that expresses GFP-luciferase throughout the parasite life cycle.
Ashley M. Vaughan,Sebastian A. Mikolajczak,Nelly Camargo,Viswanathan Lakshmanan,Mark Kennedy,Scott E. Lindner,Jessica L. Miller,Jen C. C. Hume,Stefan H. I. Kappe +8 more
TL;DR: A transgenic P. falciparum NF54 parasite that expresses a GFP-luciferase transgene throughout the life cycle is created that will accelerate testing of interventions against pre-erythrocytic life cycle stages.
Journal ArticleDOI
Household cost of malaria overdiagnosis in rural Mozambique
Jen C. C. Hume,Jen C. C. Hume,Guy Barnish,Tara D. Mangal,Luiz Armázio,Elizabeth Streat,Imelda Bates +6 more
TL;DR: An observational cohort study documenting the number and cost of repeat consultations as a result of malaria overdiagnosis at two health care providers in a rural district of Mozambique finds that investment in accurate malaria diagnosis and appropriate management at primary level is critical for improving health outcomes and reducing poverty.
Journal ArticleDOI
Two chemoattenuated PfSPZ malaria vaccines induce sterile hepatic immunity
Agnes Mwakingwe-Omari,Agnes Mwakingwe-Omari,Sara A. Healy,Jacquelyn Lane,David M. Cook,Sahand Kalhori,Charles Wyatt,Aarti Kolluri,Omely Marte-Salcedo,Alemush Imeru,Martha Nason,Lei K. Preston Church,Anita Manoj,Anusha Gunasekera,Charles Anderson,Sean C. Murphy,Sandra March,Sangeeta N. Bhatia,Eric R. James,Peter F. Billingsley,B. Kim Lee Sim,Thomas L. Richie,Irfan Zaidi,Stephen L. Hoffman,Patrick E. Duffy +36 more
TL;DR: In this article, aseptic, purified, cryopreserved, infectious Plasmodium falciparum sporozoites (PfSPZ) were inoculated under prophylactic cover with pyrimethamine (PYR) (Sanaria PfSPZ-CVac(PYR)) or chloroquine (CQ), which kill liver-stage and blood-stage parasites, respectively.
