Journal ArticleDOI
Manganese superoxide dismutase and aldehyde dehydrogenase deficiency increase mitochondrial oxidative stress and aggravate age-dependent vascular dysfunction
Philip Wenzel,Swenja Schuhmacher,Joachim Kienhöfer,Johanna Müller,Marcus Hortmann,Matthias Oelze,Eberhard Schulz,Nicolai Treiber,Toshihiro Kawamoto,Karin Scharffetter-Kochanek,Thomas Münzel,Alexander Bürkle,Markus Bachschmid,Andreas Daiber +13 more
TL;DR: The correlation between mtROS formation and acetylcholine-dependent relaxation revealed that mitochondrial radical formation significantly contributes to age-dependent endothelial dysfunction.
Journal ArticleDOI
First evidence for a crosstalk between mitochondrial and NADPH oxidase-derived reactive oxygen species in nitroglycerin-triggered vascular dysfunction.
Philip Wenzel,Hanke Mollnau,Matthias Oelze,Eberhard Schulz,Jennifer M. Dias Wickramanayake,Johanna Müller,Swenja Schuhmacher,Marcus Hortmann,Stephan Baldus,Tommaso Gori,Ralf P. Brandes,Thomas Münzel,Andreas Daiber +12 more
TL;DR: A crosstalk between mitochondrial and Nox-derived ROS with distinct mechanistic effects and sites for pharmacological intervention is suggested, suggesting a role of mitochondria for nitrate tolerance development.
Journal ArticleDOI
AT1-receptor blockade by telmisartan upregulates GTP-cyclohydrolase I and protects eNOS in diabetic rats.
Philip Wenzel,Eberhard Schulz,Matthias Oelze,Johanna Müller,Swenja Schuhmacher,Mohamed S.S. Alhamdani,Johannes Debrezion,Marcus Hortmann,Kurt Reifenberg,Ingrid Fleming,Thomas Münzel,Andreas Daiber +11 more
TL;DR: It is demonstrated for the first time that AT1-receptor blockade by telmisartan prevents downregulation of the BH4 synthase GCH-I and thereby eNOS uncoupling in experimental diabetes.
Journal ArticleDOI
ALDH-2 deficiency increases cardiovascular oxidative stress--evidence for indirect antioxidative properties.
Philip Wenzel,Johanna Müller,Sarah Zurmeyer,Swenja Schuhmacher,Eberhard Schulz,Matthias Oelze,Andrea Pautz,Toshihiro Kawamoto,Leszek Wojnowski,Hartmut Kleinert,Thomas Münzel,Andreas Daiber +11 more
TL;DR: The results suggest that ALDH-2 has an indirect antioxidative property independent of its thiol-moiety in disease states of cardiovascular oxidative stress.
Journal ArticleDOI
Chemical model systems for cellular nitros(yl)ation reactions.
Andreas Daiber,Stefan Schildknecht,Johanna Müller,Jens Kamuf,Markus Bachschmid,Volker Ullrich +5 more
TL;DR: This work proposed two new systems for nitros(yl)ation that avoid the shortcomings of previous models and developed several hypothetical scenarios to which the described (.)NO/UO2-flux model could apply, providing conditions that allow either direct electrophilic substitution at a thiolate or S-nitrosyl cation via transnitrosation from S-Nitrosoglutathione.
