Journal ArticleDOI
The mTOR inhibitor CCI-779 induces apoptosis and inhibits growth in preclinical models of primary adult human ALL
David T. Teachey,Dana A. Obzut,Dana A. Obzut,Dana A. Obzut,Jonathan Cooperman,Jonathan Cooperman,Jonathan Cooperman,Junjie Fang,Junjie Fang,Junjie Fang,Martin Carroll,Martin Carroll,Martin Carroll,John K. Choi,John K. Choi,John K. Choi,Peter J. Houghton,Peter J. Houghton,Peter J. Houghton,Valerie I. Brown,Valerie I. Brown,Valerie I. Brown,Stephan A. Grupp,Stephan A. Grupp,Stephan A. Grupp +24 more
TL;DR: It is concluded that MTIs can inhibit the growth of adult human ALL and deserve close examination as therapeutic agents against a disease that is often not curable with current therapy.
Journal ArticleDOI
Rapamycin is active against B-precursor leukemia in vitro and in vivo, an effect that is modulated by IL-7-mediated signaling.
Valerie I. Brown,Junjie Fang,Keith Alcorn,Rosalind Barr,Jenny M. Kim,Robert Wasserman,Stephan A. Grupp +6 more
TL;DR: It is found that rapamycin inhibited growth of B-precursor acute lymphoblastic leukemia lines in vitro, with evidence of apoptotic cell death, and that one of the growth signals inhibited by this class of drugs in precursor B leukemic cells may be IL-7-mediated.
Journal ArticleDOI
Thymic Stromal-Derived Lymphopoietin Induces Proliferation of Pre-B Leukemia and Antagonizes mTOR Inhibitors, Suggesting a Role for Interleukin-7Rα Signaling
Valerie I. Brown,Jessica Hulitt,Jonathan D. Fish,Cecilia Sheen,Marlo Bruno,Qing Xu,Martin Carroll,Junjie Fang,David T. Teachey,Stephan A. Grupp +9 more
TL;DR: It is shown that TSLP stimulates proliferation of pre-B ALL cell lines and partially reverses the effects of MTI on proliferation, apoptosis, and ribosomal protein S6 and 4E-BP1 phosphorylation in cell lines, with similar biological effects seen in some primary human lymphoblast samples.
Journal ArticleDOI
CD34 selection as a stem cell purging strategy for neuroblastoma: preclinical and clinical studies.
John W. Donovan,Jennifer S. Temel,Amy Zuckerman,John G. Gribben,Junjie Fang,G. Pierson,Amy A. Ross,Lisa Diller,Stephan A. Grupp,Stephan A. Grupp +9 more
TL;DR: Data show that purging of NB from PBPC specimens using CD34 selection is feasible, yielding infused products that are negative at the level of ICC but often positive at thelevel of RT-PCR.
Journal Article
Cytoplasmic μ Heavy Chain Confers Sensitivity to Dexamethasone-induced Apoptosis in Early B-lineage Acute Lymphoblastic Leukemia
Jenny M. Kim,Junjie Fang,Susan R. Rheingold,Richard Aplenc,Robert Wasserman,Stephan A. Grupp +5 more
TL;DR: A role for the pre-BCR complex is suggested in the response of ALL cells to treatment and insight is provided into the mechanism of steroid response in the treatment of pre-B ALL.
