Non-homeostatic body weight regulation through a brainstem-restricted receptor for GDF15
Jer-Yuan Hsu,Suzanne Christine Crawley,Michael Chen,Dina A. Ayupova,Darrin A. Lindhout,Higbee Jared Martin,Alan Kutach,William Joo,Zhengyu Gao,Diana Fu,Carmen To,Kalyani Mondal,Betty Chan Li,Avantika Kekatpure,Marilyn Wang,Teresa Laird,Geoffrey Horner,Jackie Chan,Michele McEntee,Manuel E. Allan +33 more
TL;DR: This work identifies glial cell-derived neurotrophic factor receptor alpha-like (GFRAL) as a brainstem-restricted receptor for growth and differentiation factor 15 (GDF15), and identifies a mechanistic basis for the non-homeostatic regulation of neural circuitry by a peripheral signal associated with tissue damage and stress.
Journal ArticleDOI
Separating Tumorigenicity from Bile Acid Regulatory Activity for Endocrine Hormone FGF19
Mei Zhou,Xueyan Wang,Van Phung,Darrin A. Lindhout,Kalyani Mondal,Jer-Yuan Hsu,Hong Yang,Mark Humphrey,Xunshan Ding,Taruna Arora,R. Marc Learned,Alex M. DePaoli,Hui Tian,Lei Ling +13 more
TL;DR: An engineered FGF19 (M70) is described that fully retains bile acid regulatory activity but does not promote HCC formation, demonstrating that regulating bile Acid metabolism is distinct and separable from tumor-promoting activity.
Journal ArticleDOI
Antidiarrhoeal activity of Cyperus rotundus.
TL;DR: The methanol extract of Cyperus rotundus rhizome, given orally at the doses of 250 and 500 mg/kg b.w., showed significant antidiarrhoeal activity in castor oil induced diarrhoea in mice.
Journal ArticleDOI
Antibody-mediated inhibition of GDF15–GFRAL activity reverses cancer cachexia in mice
Rowena Suriben,Michael Chen,Higbee Jared Martin,Julie Oeffinger,Richard Ventura,Betty Chan Li,Kalyani Mondal,Zhengyu Gao,Dina A. Ayupova,Pranali Taskar,Diana Li,Shelley R. Starck,Hung-I Harry Chen,Michele McEntee,Subhash D. Katewa,Van Phung,Marilyn Wang,Avantika Kekatpure,Damodharan Lakshminarasimhan,André O. White,Andrea Olland,Raj Haldankar,Mark J. Solloway,Jer-Yuan Hsu,Yan Wang,Jie Tang,Darrin A. Lindhout,Bernard B. Allan +27 more
TL;DR: A therapeutic antagonistic monoclonal antibody, 3P10, is reported that targets GFRAL and inhibits RET signaling by preventing the GDF15-driven interaction of RET with GFRal on the cell surface and reverses excessive lipid oxidation in tumor-bearing mice and prevents cancer cachexia, even under calorie-restricted conditions.
