MKS and NPHP modules cooperate to establish basal body/transition zone membrane associations and ciliary gate function during ciliogenesis
Corey L. Williams,Chunmei Li,Katarzyna Kida,Peter N. Inglis,Swetha Mohan,Lucie Semenec,Nathan J. Bialas,Rachel M. Stupay,Nansheng Chen,Oliver E. Blacque,Bradley K. Yoder,Michel R. Leroux +11 more
TL;DR: Eight proteins, defects in which are associated with Meckel-Gruber syndrome and nephronophthisis ciliopathies, work together as two functional modules at the transition zone to establish basal body/transition zone connections with the membrane and barricade entry of non-ciliary components into this organelle.
Journal ArticleDOI
Joubert syndrome Arl13b functions at ciliary membranes and stabilizes protein transport in Caenorhabditis elegans
Sebiha Cevik,Yuji Hori,Oktay I. Kaplan,Katarzyna Kida,Tiina Toivenon,Christian Foley-Fisher,David C. Cottell,Toshiaki Katada,Kenji Kontani,Oliver E. Blacque +9 more
TL;DR: The small GTPase Arl13b regulates ciliary transmembrane protein localizations and anterograde IFT assembly stability.
Journal ArticleDOI
Bistability in the Rac1, PAK, and RhoA Signaling Network Drives Actin Cytoskeleton Dynamics and Cell Motility Switches
Kate M. Byrne,Naser Monsefi,John C. Dawson,Andrea Degasperi,Jimi-Carlo Bukowski-Wills,Natalia Volinsky,Maciej Dobrzyński,Marc R. Birtwistle,Mikhail A Tsyganov,Anatoly Kiyatkin,Katarzyna Kida,Andrew J. Finch,Neil O. Carragher,Walter Kolch,Lan K. Nguyen,Lan K. Nguyen,Alex von Kriegsheim,Boris N. Kholodenko +17 more
TL;DR: It is shown that a network containing Rac1, RhoA, and PAK family kinases can produce bistable, switch-like responses to a graded PAK inhibition, and that PAK is a critical component in the Rac1-RhoA inhibitory crosstalk that governs bistables GTPase activity, cell morphology, and cell migration switches.
Journal ArticleDOI
Active Transport and Diffusion Barriers Restrict Joubert Syndrome-Associated ARL13B/ARL-13 to an Inv-like Ciliary Membrane Subdomain
Sebiha Cevik,Anna A. W. M. Sanders,Erwin van Wijk,Karsten Boldt,Lara Clarke,Jeroen van Reeuwijk,Yuji Hori,Nicola Horn,Lisette Hetterschijt,Anita Wdowicz,Andrea Mullins,Katarzyna Kida,Oktay I. Kaplan,Oktay I. Kaplan,Sylvia E. C. van Beersum,Ka Man Wu,Stef J.F. Letteboer,Dorus A. Mans,Toshiaki Katada,Kenji Kontani,Marius Ueffing,Marius Ueffing,Ronald Roepman,Hannie Kremer,Oliver E. Blacque +24 more
TL;DR: The transport mechanisms underlying compartmentalization of JS-associated ARL13B/ARL-13, which was previously found is restricted at proximal ciliary membranes are investigated, reveal distinct requirements for sequence motifs, IFT and ciliopathy modules in defining an ARL- 13 subciliary membrane compartment.
Journal ArticleDOI
Endocytosis Genes Facilitate Protein and Membrane Transport in C. elegans Sensory Cilia
Oktay I. Kaplan,David B. Doroquez,Sebiha Cevik,Rachel V. Bowie,Lara Clarke,Anna A. W. M. Sanders,Katarzyna Kida,Joshua Z. Rappoport,Piali Sengupta,Oliver E. Blacque +9 more
TL;DR: These data implicate C. elegans endocytosis proteins localized at the ciliary base in regulating ciliary and periciliary membrane volume and suggest that membrane retrieval from these compartments is counterbalanced by BBS-8 and RAB-8-mediated membrane delivery.
