A cytomegalovirus-encoded mitochondria-localized inhibitor of apoptosis structurally unrelated to Bcl-2
Victor S. Goldmacher,Laura M. Bartle,Anna Skaletskaya,Cheryl A. Dionne,Nancy Kedersha,Carol A. Vater,Jia-wen Han,Robert J. Lutz,Shinya Watanabe,Ellen Cahir McFarland,Elliott Kieff,Edward S. Mocarski,Thomas Chittenden +12 more
TL;DR: Human cytomegalovirus (CMV), a herpesvirus that causes congenital disease and opportunistic infections in immunocompromised individuals, encodes functions that facilitate efficient viral propagation by altering host cell behavior and encodes a mitochondria-localized inhibitor of apoptosis, denoted vMIA, capable of suppressing apoptosis induced by diverse stimuli.
Journal ArticleDOI
A cytomegalovirus-encoded inhibitor of apoptosis that suppresses caspase-8 activation.
Anna Skaletskaya,Laura M. Bartle,Thomas Chittenden,A. Louise McCormick,Edward S. Mocarski,Victor S. Goldmacher +5 more
TL;DR: A human cytomegalovirus cell-death suppressor, denoted vICA, encoded by the viral UL36 gene is identified, which inhibits Fas-mediated apoptosis by binding to the pro-domain of caspase-8 and preventing its activation.
Journal ArticleDOI
SAR650984, a novel humanized CD38-targeting antibody, demonstrates potent antitumor activity in models of multiple myeloma and other CD38+ hematologic malignancies.
Jutta Deckert,Marie-Cécile Wetzel,Laura M. Bartle,Anna Skaletskaya,Victor S. Goldmacher,François Vallée,Qing Zhou-Liu,Paul Ferrari,Stéphanie Pouzieux,Charlotte Lahoute,Charles Dumontet,Adriana Plesa,Marielle Chiron,Pascale Lejeune,Thomas Chittenden,Peter U. Park,Véronique Blanc +16 more
TL;DR: These results validate CD38 as a therapeutic target and support the current evaluation of this unique CD38-targeting functional antibody in phase I clinical trials in patients with CD38+ B-cell malignancies.
Journal ArticleDOI
The adenine nucleotide translocator: a target of nitric oxide, peroxynitrite, and 4-hydroxynonenal
Helena L. A. Vieira,Anne Sophie Belzacq,Delphine Haouzi,Francesca Bernassola,Isabel Cohen,Etienne Jacotot,Karine F. Ferri,Chahrazed El Hamel,Laura M. Bartle,Gerry Melino,Catherine Brenner,Catherine Brenner,Victor S. Goldmacher,Guido Kroemer +13 more
TL;DR: In vitro data suggest that NO, peroxynitrite, and HNE may directly act on ANT to induce mitochondrial membrane permeabilization and apoptosis.
Journal ArticleDOI
Cytomegalovirus cell death suppressor vMIA blocks Bax- but not Bak-mediated apoptosis by binding and sequestering Bax at mitochondria
Damien Arnoult,Laura M. Bartle,Anna Skaletskaya,Delphine Poncet,Naoufal Zamzami,Peter U. Park,Juanita C. Sharpe,Richard J. Youle,Victor S. Goldmacher +8 more
TL;DR: It appears that vMIA is capable of suppressing apoptosis in cells in which this pathway is dominated by Bax, but not in cells where Bak also plays a role, and does not seem to either specifically associate with Bak or suppress Bak-mediated mitochondrial membrane permeabilization.
