Journal ArticleDOI
Anti-inflammatory and immunomodulatory properties of α1-antitrypsin without inhibition of elastase
Danny Jonigk,Mariam M. Al-Omari,Lavinia Maegel,Meike Müller,Nicole Izykowski,Jaewoo Hong,Kwangwon Hong,Soohyun Kim,Martina Dorsch,Ravi Mahadeva,Florian Laenger,Hans Kreipe,Armin Braun,Galit Shahaf,Eli C. Lewis,Tobias Welte,Charles A. Dinarello,Sabina Janciauskiene +17 more
TL;DR: Consistently, in vivo and in vitro, rAAT reduced inflammatory responses at concentrations 40- to 100-fold lower than native plasma-derived AAT, providing evidence that the anti-inflammatory and immunomodulatory properties of AAT can be independent of elastase inhibition.
Journal ArticleDOI
PPARγ agonist pioglitazone reverses pulmonary hypertension and prevents right heart failure via fatty acid oxidation
Ekaterina Legchenko,Philippe Chouvarine,Paul Borchert,Angeles Fernandez-Gonzalez,Erin Snay,Martin Meier,Lavinia Maegel,S. Alex Mitsialis,Eva A. Rog-Zielinska,Stella Kourembanas,Danny Jonigk,Georg Hansmann +11 more
TL;DR: It is shown that activation of the peroxisome proliferator–activated receptor γ (PPARγ) via pioglitazone treatment protects against heart failure in the Sugen hypoxia rat model of pulmonary arterial hypertension and suggests that targeting PPARγ activation to restore fatty acid oxidation could be therapeutic for pulmonary hypertension and other diseases with altered lipid metabolism.
Journal ArticleDOI
Plexiform Lesions in Pulmonary Arterial Hypertension: Composition, Architecture, and Microenvironment
Danny Jonigk,Heiko Golpon,Clemens L. Bockmeyer,Lavinia Maegel,Marius M. Hoeper,Jens Gottlieb,Nils Nickel,Kais Hussein,Ulrich A. Maus,Ulrich Lehmann,Sabina Janciauskiene,Tobias Welte,Axel Haverich,Johanna Rische,Hans Kreipe,Florian Laenger +15 more
TL;DR: PLs show a distinct cellular composition and microenvironment, which contribute to the plexiform phenotype and set them apart from other processes of vascular remodeling in patients with PAH.
Journal ArticleDOI
Circulating angiopoietins in idiopathic pulmonary arterial hypertension
Philipp Kümpers,Nils Nickel,Alexander Lukasz,Heiko Golpon,Volker Westerkamp,Karen M. Olsson,Danny Jonigk,Lavinia Maegel,Clemens L. Bockmeyer,Sascha David,Sascha David,Marius M. Hoeper +11 more
TL;DR: Ang-2 may be involved in the pathogenesis of IPAH, and plasma Ang-2 might serve as a promising new biomarker of disease severity and response to treatment in patients with IPAH.
Journal ArticleDOI
Notch1 signalling regulates endothelial proliferation and apoptosis in pulmonary arterial hypertension.
Swati Dabral,Xia Tian,Baktybek Kojonazarov,Rajkumar Savai,Hossein Ardeschir Ghofrani,Norbert Weissmann,Monica Florio,Jan Sun,Danny Jonigk,Lavinia Maegel,Friedrich Grimminger,Werner Seeger,Soni Savai Pullamsetti,Ralph T. Schermuly +13 more
TL;DR: It is concluded that Notch1 plays a critical role in PAH and Notch inhibitors may be a promising therapeutic option for PAH.
