The 2019 coronavirus (SARS-CoV-2) surface protein (Spike) S1 Receptor Binding Domain undergoes conformational change upon heparin binding
Courtney J. Mycroft-West,Dunhao Su,Stefano Elli,Yong Li,Scott E. Guimond,Gavin J. Miller,Jeremy E. Turnbull,Edwin A. Yates,Marco Guerrini,David G. Fernig,Marcelo A. Lima,Mark A. Skidmore +11 more
TL;DR: The data demonstrate an interaction between the recombinant surface receptor binding domain and the polysaccharide that has implications for the rapid development of a first-line therapeutic by repurposing heparin and for next-generation, tailor-made, GAG-based antivirals.
Journal ArticleDOI
Heparin Inhibits Cellular Invasion by SARS-CoV-2: Structural Dependence of the Interaction of the Spike S1 Receptor-Binding Domain with Heparin.
Courtney J. Mycroft-West,Dunhao Su,Isabel Pagani,Timothy R. Rudd,Stefano Elli,Neha S. Gandhi,Scott E. Guimond,Gavin J. Miller,Maria Cecília Zorél Meneghetti,Helena B. Nader,Yong Li,Quentin M. Nunes,Patricia Procter,Nicasio Mancini,Massimo Clementi,Antonella Bisio,Nicholas R. Forsyth,Vito Ferro,Jeremy E. Turnbull,Marco Guerrini,David G. Fernig,Elisa Vicenzi,Edwin A. Yates,Edwin A. Yates,Marcelo A. Lima,Mark A. Skidmore,Mark A. Skidmore +26 more
TL;DR: In this paper, surface plasmon resonance and circular dichroism spectroscopy were used to investigate the structural basis of heparin and glycosaminoglycan heparan sulfate (HS) binding to the spike protein receptor-binding domain (S1 RBD) of SARS-CoV-2.
Journal ArticleDOI
Heparin prevents Zika virus induced-cytopathic effects in human neural progenitor cells
Silvia Ghezzi,Lynsay C. Cooper,Alicia Rubio,Isabel Pagani,Maria Rosaria Capobianchi,Giuseppe Ippolito,Julien Pelletier,Maria Cecília Zorél Meneghetti,Marcelo A. Lima,Mark A. Skidmore,Vania Broccoli,Edwin A. Yates,Edwin A. Yates,Edwin A. Yates,Elisa Vicenzi +14 more
TL;DR: It is shown that heparin, a widely used anticoagulant, while exerting a modest inhibitory effect on Zika Virus replication, fully prevents virus‐induced cell death of human neural progenitor cells (NPCs).

Lima,Mark A. Skidmore,Mark A. Skidmore +24 more
TL;DR: Heparin prevents infection by a range of viruses if added exogenously, including S-associated coronavirus strain HSR1, and inhibits invasion by SARS-CoV-2 and other members of the Coronaviridae.
Journal ArticleDOI
Diversification of the Structural Determinants of Fibroblast Growth Factor-Heparin Interactions IMPLICATIONS FOR BINDING SPECIFICITY
Ruoyan Xu,Alessandro Ori,Timothy R. Rudd,Katarzyna A. Uniewicz,Yassir A. Ahmed,Scott E. Guimond,Mark A. Skidmore,Mark A. Skidmore,Giuliano Siligardi,Edwin A. Yates,David G. Fernig +10 more
TL;DR: Differential scanning fluorimetry identified clear preferences in these FGFs for distinct structural features in the polysaccharide, suggesting that the differences in heparin-binding sites in both the protein and the sugar are greatest between subfamilies and may be more restricted within a FGF subfamily in accord with the known conservation of function within FGFSubfamilies.
