Journal ArticleDOI
Generation of high-affinity human antibodies by combining donor-derived and synthetic complementarity-determining-region diversity
Rene Hoet,Edward H. Cohen,Rachel Baribault Kent,Kristin L. Rookey,Sonia Schoonbroodt,Shannon Hogan,Louise Rem,Nicolas Frans,Marc Daukandt,Henk Pieters,Rob van Hegelsom,Nicole Coolen-van Neer,Nastri Horacio G,Isaac J. Rondon,Jennifer A. Leeds,Simon E. Hufton,Lili Huang,Irina Kashin,Mary Devlin,Guannan Kuang,Mieke Steukers,Malini Viswanathan,Andrew E. Nixon,Daniel J. Sexton,Hennie R. Hoogenboom,Robert Charles Ladner +25 more
TL;DR: The construction of human Fab libraries having a unique combination of immunoglobulin sequences captured from human donors and synthetic diversity in key antigen contact sites in heavy-chain complementarity-determining regions 1 and 2 are reported.
Journal ArticleDOI
Affinity maturation of Fab antibody fragments by fluorescent-activated cell sorting of yeast-displayed libraries.
Twan van den Beucken,Henk Pieters,Mieke Steukers,Marcel van der Vaart,Robert Charles Ladner,Hennie R. Hoogenboom,Simon E. Hufton +6 more
TL;DR: The ability to efficiently affinity mature Fab antibodies is an important component of the antibody development pipeline and it is shown that yeast display is an efficient method for this purpose.
Journal ArticleDOI
Construction and diversification of yeast cell surface displayed libraries by yeast mating: application to the affinity maturation of Fab antibody fragments.
Lydia Blaise,Anita Wehnert,Mieke Steukers,Twan van den Beucken,Hennie R. Hoogenboom,Simon E. Hufton +5 more
TL;DR: Using error-prone polymerase chain reaction, a naive human Fab yeast display library of over 10(9) clones is generated and independent repertoires of heavy chain (HC) and light chain (LC) can be constructed in haploid yeast strains of opposite mating type.
Journal ArticleDOI
Engineering Antibody Heavy Chain CDR3 to Create a Phage Display Fab Library Rich in Antibodies That Bind Charged Carbohydrates
Sonia Schoonbroodt,Mieke Steukers,Malini Viswanathan,Nicolas Frans,Marie Timmermans,Anita Wehnert,Minh Nguyen,Robert Charles Ladner,Rene Hoet +8 more
TL;DR: The design of an Ab repertoire enriched in Abs binding to small charged carbohydrates and the construction of a human Fab phagemid library, “FAB-CCHO,” are reported.
Journal ArticleDOI
Rapid kinetic-based screening of human Fab fragments.
Mieke Steukers,Jean-Michel Schaus,Reinoud van Gool,Anne Hoyoux,Pascale Richalet,Daniel J. Sexton,Andrew E. Nixon,Marc Vanhove +7 more
TL;DR: A Biacore assay that allows isolated clones expressed as soluble Fab fragments in E. coli to be screened and ranked based on their affinity against the target and takes advantage of the ability to measure Fab concentrations in crude bacterial extracts inBiacore using very high density Protein A chips.
