Journal ArticleDOI

The repertoire of λ light chains causing predominant amyloid heart involvement and identification of a preferentially involved germline gene, IGLV1-44.

TL;DR: The repertoire of variable region germline genes of λ LC preferentially targeting the heart was characterized and a single gene, IGVL1-44, was found associated with a 5-fold increase in the odds of dominant heart involvement (after adjusting for confounders in a multivariable logistic model).
ComponentDOI

Cryo-EM structure of cardiac amyloid fibrils from an immunoglobulin light chain AL amyloidosis patient.

TL;DR: The authors' data shed light on the architecture of LC amyloids, correlate amino acid sequences with fibril assembly, providing the grounds for development of innovative medicines, and highlight structural motifs that may be common to misfolding-prone LCs.
Journal ArticleDOI

The amyloidogenic light chain is a stressor that sensitizes plasma cells to proteasome inhibitor toxicity

TL;DR: An unprecedented intrinsic sensitivity of AL PCs to PIs is revealed, even higher than that of MM PCs, associated with distinctive organellar features and expression patterns indicative of cellular stress, and stress-responsive pathways as novel potential therapeutic targets are identified.
