IL4I1 Is a Metabolic Immune Checkpoint that Activates the AHR and Promotes Tumor Progression
Ahmed Sadik,Luis F. Somarribas Patterson,Selcen Öztürk,Soumya R. Mohapatra,Verena Panitz,Philipp F Secker,Pauline Pfänder,Stefanie Loth,Heba Salem,Mirja Tamara Prentzell,Bianca Berdel,Murat Iskar,Erik Faessler,Friederike Reuter,Isabelle Kirst,Verena Kalter,Kathrin I. Foerster,Evelyn Jäger,Carina Ramallo Guevara,Mansour Sobeh,Thomas Hielscher,Gernot Poschet,Annekathrin Reinhardt,Jessica C. Hassel,Marc Zapatka,Udo Hahn,Andreas von Deimling,Carsten Hopf,Rita Schlichting,Beate I. Escher,Jürgen Burhenne,Walter E. Haefeli,Naveed Ishaque,Alexander Böhme,Sascha Schäuble,Kathrin Thedieck,Saskia Trump,Martina Seiffert,Christiane A. Opitz +38 more
TL;DR: A pan-tissue AHR signature revealed that across 32 tumor entities, interleukin-4-induced-1 (IL4I1) associates more frequently with AHR activity than IDO1 or TDO2, hitherto recognized as the main Trp-catabolic enzymes.
Posted ContentDOI
Comparative Assessment of Quantification Methods for Tumor Tissue Phosphoproteomics
Yang Zhang,Benjamin Dreyer,Natalia Govorukhina,Alexander Martin Heberle,Sasa Koncarevic,Christoph Krisp,Christiane A. Opitz,Pauline Pfänder,Rainer Bischoff,Hartmut Schlüter,Marcel Kwiatkowski,Kathrin Thedieck,Peter Horvatovich +12 more
TL;DR: This study compared label-free quantification, spike-in-SILAC and TMT isobaric tandem mass tags technology for quantitative phosphosite profiling in tumor tissue and found TMT offered the lowest accuracy and the highest precision and robustness towards different phosphosite abundances and matrices.
Journal ArticleDOI
Device-Controlled Microcondensation for Spatially Confined On-Tissue Digests in MALDI Imaging of N-Glycans
Annabelle Fülöp,Christian Marsching,Frederik Barka,Yasemin Ucal,Pauline Pfänder,Christiane A. Opitz,Günes Barka,Carsten Hopf +7 more
TL;DR: In this paper , a device-controlled workflow for on-tissue enzymatic digestion of N-glycans was developed for mass spectrometry imaging (MSI) and spatialomic analysis of tissue proteins.
Posted ContentDOI
Tryptophan stress activates EGFR-RAS-signaling to MTORC1 and p38/MAPK to sustain translation and AHR-dependent autophagy
Pauline Pfänder,Lucas Hensen,Patricia Razquin Navas,M J Solvay,Mirja Tamara Prentzell,Ahmed Sadik,Alexander Martin Heberle,Sophie Seifert,Leon Regin,Tobias Bausbacher,Anna Sophia Egger,Madlen Hotze,Tobias Kipura,Bianca Berdel,I. Korolchuk,Shad Arif Mohammed,Phillipp Sievers,Felix Sahm,Hartmut Schlüter,Andreas Koeberle,Carsten Hopf,M. Kwiatkowski,Christine Sers,Benoît Van den Eynde,Christiane A. Opitz,Kathrin Thedieck +42 more
TL;DR: In this article , the authors show that Trp restriction not only inhibits MTORC1 and AHR, but also promotes autophagy and ceramide biosynthesis in glioblastoma patients.
