Journal ArticleDOI
A systems study reveals concurrent activation of AMPK and mTOR by amino acids
Piero Dalle Pezze,Stefanie Ruf,Stefanie Ruf,Annika G Sonntag,Miriam Langelaar-Makkinje,Philip Hall,Alexander Martin Heberle,Patricia Razquin Navas,Patricia Razquin Navas,Karen van Eunen,Regine C Tölle,Jennifer J. Schwarz,Heike Wiese,Bettina Warscheid,Jana Deitersen,Björn Stork,Erik Fäßler,Sascha Schäuble,Udo Hahn,Peter Horvatovich,Daryl P. Shanley,Kathrin Thedieck,Kathrin Thedieck +22 more
TL;DR: It is found that aa acutely activate AMPK concurrently with mTOR, and it is shown that AMPK under aa sufficiency acts to sustain autophagy, which may be required to maintain protein homoeostasis and deliver metabolite intermediates for biosynthetic processes.
Journal ArticleDOI
IL4I1 Is a Metabolic Immune Checkpoint that Activates the AHR and Promotes Tumor Progression
Ahmed Sadik,Luis F. Somarribas Patterson,Selcen Öztürk,Soumya R. Mohapatra,Verena Panitz,Philipp F Secker,Pauline Pfänder,Stefanie Loth,Heba Salem,Mirja Tamara Prentzell,Bianca Berdel,Murat Iskar,Erik Faessler,Friederike Reuter,Isabelle Kirst,Verena Kalter,Kathrin I. Foerster,Evelyn Jäger,Carina Ramallo Guevara,Mansour Sobeh,Thomas Hielscher,Gernot Poschet,Annekathrin Reinhardt,Jessica C. Hassel,Marc Zapatka,Udo Hahn,Andreas von Deimling,Carsten Hopf,Rita Schlichting,Beate I. Escher,Jürgen Burhenne,Walter E. Haefeli,Naveed Ishaque,Alexander Böhme,Sascha Schäuble,Kathrin Thedieck,Saskia Trump,Martina Seiffert,Christiane A. Opitz +38 more
TL;DR: A pan-tissue AHR signature revealed that across 32 tumor entities, interleukin-4-induced-1 (IL4I1) associates more frequently with AHR activity than IDO1 or TDO2, hitherto recognized as the main Trp-catabolic enzymes.
Journal ArticleDOI
Metabolic Adaptation Establishes Disease Tolerance to Sepsis
Sebastian Weis,Ana Rita Carlos,Maria Raquel Moita,Sumnima Singh,Birte Blankenhaus,Silvia Cardoso,Rasmus Larsen,Sofia Rebelo,Sascha Schäuble,Laura del Barrio,Gilles Mithieux,Fabienne Rajas,Sandro Lindig,Michael Bauer,Miguel P. Soares +14 more
TL;DR: It is demonstrated that induction of the iron-sequestering ferritin H chain in response to polymicrobial infections is critical to establish disease tolerance to sepsis and a crosstalk between adaptive responses controlling iron and glucose metabolism is required to maintain blood glucose within a physiologic range compatible with host survival.
Journal ArticleDOI
Metabolic costs of amino acid and protein production in Escherichia coli.
TL;DR: It is shown that the biosynthesis of a large number of amino acids from glucose and particularly from glycerol provides a surplus of energy, which can be used to balance the high energetic cost of amino acid polymerization.
Journal ArticleDOI
Transcriptomic alterations during ageing reflect the shift from cancer to degenerative diseases in the elderly.
Peer Aramillo Irizar,Sascha Schäuble,Daniela Esser,Marco Groth,Christiane Frahm,Steffen Priebe,Mario Baumgart,Nils Hartmann,Shiva Marthandan,Uwe Menzel,Julia J. Müller,Silvio Schmidt,Volker Ast,Amke Caliebe,Rainer König,Michael Krawczak,Michael Ristow,Stefan Schuster,Alessandro Cellerino,Alessandro Cellerino,Stephan Diekmann,Christoph Englert,Christoph Englert,Peter Hemmerich,Jürgen Sühnel,Reinhard Guthke,Otto W. Witte,Matthias Platzer,Eytan Ruppin,Christoph Kaleta +29 more
TL;DR: It is shown that in concordance with this shift, conserved transcriptional alterations during ageing across four vertebrates align with degenerative diseases but are opposite to those in cancer.
