Biodegradable polymeric nanoparticles based drug delivery systems

Nanoparticle (NP) drug delivery systems (5−250 nm) have the potential to improve current disease therapies because of their ability to overcome multiple biological barriers and releasing a therapeutic load in the optimal dosage range. Rapid clearance of circulating nanoparticles during systemic delivery is a critical issue for these systems and has made it necessary to understand the factors affecting particle biodistribution and blood circulation half-life. In this review, we discuss the factors which can influence nanoparticle blood residence time and organ specific accumulation. These factors include interactions with biological barriers and tunable nanoparticle parameters, such as composition, size, core properties, surface modifications (pegylation and surface charge), and finally, targeting ligand functionalization. Factors Affecting the Clearance and Biodistribution of Polymeric Nanoparticles

PEGylation as a strategy for improving nanoparticle-based drug and gene delivery

Nanocapsules are submicroscopic colloidal drug carrier systems composed of an oily or an aqueous core surrounded by a thin polymer membrane. Two technologies can be used to obtain such nanocapsules: the interfacial polymerization of a monomer or the interfacial nanodeposition of a preformed polymer. This article is an extended review of these nanocapsule technologies and their applications for the treatment of various diseases (including cancer and infections).

Cet article fait le point sur les aspects physicochimiques et les applications therapeutiques des nanocapsules en tant que systeme de delivrance de medicaments. Nous nous sommes tout d'abord interesses aux methodes de preparation des nanocapsules et aux moyens mis en oeuvre pour caracteriser ces systemes H/E (taille, surface, densite, liberation, stabilite) avant d'etudier l'influence de l'encapsulation de nombreux principes actifs sur leur activite biologique. Parmi les raisons invoquees pour justifier l'utilisation therapeutique des nanocapsules, on peut citer la possibilite de solubiliser des principes actifs peu solubles dans l'eau, celle de proteger les muqueuses d 'une toxicite locale de certains principes actifs, celle d'augmenter la permeabilite des muqueuses aux molecules actives ou celle de prolonger leur circulation dans la circulation sanguine. Depuis le milieu des annees 1980, de nombreuses molecules actives ont ete encapsulees et ont permis d'ameliorer leur activite therapeutique ou de reduire leurs effets secondaires: parmi elles, citons les anti-inflammatoires, les anticancereux, les immunostimulants, les anti-infectieux, les antiglaucomateux et meme certains peptides.
