Journal ArticleDOI
Bone recognition mechanism of porcine osteocalcin from crystal structure.
TL;DR: The X-ray crystal structure of porcine osteocalcin is presented at 2.0 Å resolution, which reveals a negatively charged protein surface that coordinates five calcium ions in a spatial orientation that is complementary to calcium ion in a hydroxyapatite crystal lattice.
Journal ArticleDOI
A soluble α-synuclein construct forms a dynamic tetramer
Wei Wang,Iva Perovic,Johnathan Chittuluru,Alice Kaganovich,Linh T.T. Nguyen,Linh T.T. Nguyen,Jingling Liao,Jingling Liao,Jared R. Auclair,Derrick E. Johnson,Derrick E. Johnson,Anuradha Landeru,Anuradha Landeru,Alana K. Simorellis,Shulin Ju,Mark R. Cookson,Francisco J. Asturias,Jeffrey N. Agar,Brian N. Webb,ChulHee Kang,Dagmar Ringe,Dagmar Ringe,Gregory A. Petsko,Gregory A. Petsko,Thomas C. Pochapsky,Quyen Q. Hoang +25 more
TL;DR: A dynamic model for the tetramer structure is proposed, based on expected close association of the amphipathic central helices observed in the previously described micelle-associated “hairpin” structure of α-synuclein, which indicates that a symmetric oligomer is present, with three- or fourfold symmetry.
Journal ArticleDOI
Caspase-1 causes truncation and aggregation of the Parkinson’s disease-associated protein α-synuclein
Wei Wang,Linh T.T. Nguyen,Christopher Burlak,Fariba Chegini,Feng Guo,Tim Chataway,Shulin Ju,Oriana S. Fisher,David Miller,Debajyoti Datta,Fang Wu,Chun Xiang Wu,Anuradha Landeru,James A. Wells,Mark R. Cookson,Matthew B. Boxer,Craig J. Thomas,Wei Ping Gai,Dagmar Ringe,Gregory A. Petsko,Gregory A. Petsko,Gregory A. Petsko,Quyen Q. Hoang +22 more
TL;DR: In this paper, the role of inflammation in the aggregation of α-synuclein (aSyn) is investigated in a neuronal cell model of Parkinson's disease (PD) and it is shown that activation of the inflammasome with known stimuli results in aggregation of aSyn.
Journal ArticleDOI
Structural basis for conformational plasticity of the Parkinson's disease-associated ubiquitin hydrolase UCH-L1
Chittaranjan Das,Quyen Q. Hoang,Cheryl A. Kreinbring,Sarah J. Luchansky,Robin K. Meray,Soumya S. Ray,Peter T. Lansbury,Dagmar Ringe,Dagmar Ringe,Gregory A. Petsko,Gregory A. Petsko +10 more
TL;DR: The three-dimensional structure of human UCH-L1 is determined at 2.4-A resolution by x-ray crystallography, and the overall fold resembles that of other ubiquitin hydrolases, including UCH- L3, but there are a number of significant differences.
Caspase-1 causes truncation and aggregation of the Parkinson's disease-associated protein α-synuclein
Wei Wang,Linh T.T. Nguyen,Christopher Burlak,Fariba Chegini,Feng Guo,Tim Chataway,Shulin Ju,Oriana S. Fisher,David Miller,Debajyoti Datta,Fang Wu,Chun Xiang Wu,Anuradha Landeru,James A. Wells,Mark R. Cookson,Matthew B. Boxer,Craig J. Thomas,Wei Ping Gai,Dagmar Ringe,Gregory A. Petsko,Gregory A. Petsko,Gregory A. Petsko,Quyen Q. Hoang +22 more
TL;DR: It is reported that inflammation can play a role in causing the aggregation of wild type aSyn, and perhaps in the pathogenesis of sporadic PD as well.
