Immune evasion in cancer: Mechanistic basis and therapeutic strategies

Obesity is associated with low-grade chronic inflammation and intestinal dysbiosis. Ganoderma lucidum is a medicinal mushroom used in traditional Chinese medicine with putative anti-diabetic effects. Here, we show that a water extract of Ganoderma lucidum mycelium (WEGL) reduces body weight, inflammation and insulin resistance in mice fed a high-fat diet (HFD). Our data indicate that WEGL not only reverses HFD-induced gut dysbiosis-as indicated by the decreased Firmicutes-to-Bacteroidetes ratios and endotoxin-bearing Proteobacteria levels-but also maintains intestinal barrier integrity and reduces metabolic endotoxemia. The anti-obesity and microbiota-modulating effects are transmissible via horizontal faeces transfer from WEGL-treated mice to HFD-fed mice. We further show that high molecular weight polysaccharides (>300 kDa) isolated from the WEGL extract produce similar anti-obesity and microbiota-modulating effects. Our results indicate that G. lucidum and its high molecular weight polysaccharides may be used as prebiotic agents to prevent gut dysbiosis and obesity-related metabolic disorders in obese individuals.

Biological activities and pharmaceutical applications of polysaccharide from natural resources: A review

Mushrooms have been consumed since earliest history; ancient Greeks believed that mushrooms provided strength for warriors in battle, and the Romans perceived them as the “Food of the Gods.” For centuries, the Chinese culture has treasured mushrooms as a health food, an “elixir of life.” They have been part of the human culture for thousands of years and have considerable interest in the most important civilizations in history because of their sensory characteristics; they have been recognized for their attractive culinary attributes. Nowadays, mushrooms are popular valuable foods because they are low in calories, carbohydrates, fat, and sodium: also, they are cholesterol-free. Besides, mushrooms provide important nutrients, including selenium, potassium, riboflavin, niacin, vitamin D, proteins, and fiber. All together with a long history as food source, mushrooms are important for their healing capacities and properties in traditional medicine. It has reported beneficial effects for health and treatment of some diseases. Many nutraceutical properties are described in mushrooms, such as prevention or treatment of Parkinson, Alzheimer, hypertension, and high risk of stroke. They are also utilized to reduce the likelihood of cancer invasion and metastasis due to antitumoral attributes. Mushrooms act as antibacterial, immune system enhancer and cholesterol lowering agents; additionally, they are important sources of bioactive compounds. As a result of these properties, some mushroom extracts are used to promote human health and are found as dietary supplements.

Anticancer polysaccharides from natural resources: A review of recent research

Ganoderma lucidum (Ling Zhi) is a basidiomycete white rot macrofungus which has been used extensively as "the mushroom of immortality" in China, Japan, Korea and other Asian countries for 2000 years. A great deal of work has been carried out on therapeutic potential of Ganoderma lucidum. The basidiocarp, mycelia and spores of Ganoderma lucidum contain approximately 400 different bioactive compounds, which mainly include triterpenoids, polysaccharides, nucleotides, sterols, steroids, fatty acids, proteins/peptides and trace elements which has been reported to have a number of pharmacological effects including immunomodulation, anti-atherosclerotic, anti-inflammatory, analgesic, chemo-preventive, antitumor, chemo and radio protective, sleep promoting, antibacterial, antiviral (including anti-HIV), hypolipidemic, anti-fibrotic, hepatoprotective, anti-diabetic, anti-androgenic, anti-angiogenic, anti-herpetic, antioxidative and radical-scavenging, anti-aging, hypoglycemic, estrogenic activity and anti-ulcer properties. Ganoderma lucidum has now become recognized as an alternative adjuvant in the treatment of leukemia, carcinoma, hepatitis and diabetes. The macrofungus is very rare in nature rather not sufficient for commercial exploitation for vital therapeutic emergencies, therefore, the cultivation on solid substrates, stationary liquid medium or by submerged cultivation has become an essential aspect to meet the driving force towards the increasing demands in the international market. Present review focuses on the pharmacological aspects, cultivation methods and bioactive metabolites playing a significant role in various therapeutic applications.

Ganoderma lucidum: a potent pharmacological macrofungus.

Lentinus edodes is the first medicinal macrofungus to enter the realm of modern biotechnology. It is the second most popular edible mushroom in the global market which is attributed not only to its nutritional value but also to possible potential for therapeutic applications. Lentinus edodes is used medicinally for diseases involving depressed immune function (including AIDS), cancer, environmental allergies, fungal infection, frequent flu and colds, bronchial inflammation, heart disease, hyperlipidemia (including high blood cholesterol), hypertension, infectious disease, diabetes, hepatitis and regulating urinary inconsistencies. It is the source of several well-studied preparations with proven pharmacological properties, especially the polysaccharide lentinan, eritadenine, shiitake mushroom mycelium, and culture media extracts (LEM, LAP and KS-2). Antibiotic, anti-carcinogenic and antiviral compounds have been isolated intracellularly (fruiting body and mycelia) and extracellularly (culture media). Some of these substances were lentinan, lectins and eritadenine. The aim of this review is to discuss the therapeutic applications of this macrofungus. The potential of this macrofungus is unquestionable in the most important areas of applied biotechnology.

Lentinus edodes: A Macrofungus with Pharmacological Activities

The production of biodiesel by transesterification employing acid or base catalyst has been industrially accepted for its high conversion and reaction rates. Downstream processing costs and environmental problems associated with biodiesel production and byproducts recovery have led to the search for alternative production methods. Recently, enzymatic transesterification involving lipases has attracted attention for biodiesel production as it produces high purity product and enables easy separation from the byproduct, glycerol. The use of immobilized lipases and immobilized whole cells may lower the overall cost, while presenting less downstream processing problems, to biodiesel production. The present review gives an overview on biodiesel production technology and analyzes the factors/methods of enzymatic approach reported in the literature and also suggests suitable method on the basis of evidence for industrial production of biodiesel.

Biodiesel production with special emphasis on lipase-catalyzed transesterification

Microbial preparation of herbicide is defined as bioherbicide that can control the weed. In this approach, indigenous plant pathogens isolated from weeds are cultured to produce the large numbers of infective propagules which are applied at a rate that will cause high levels of infection leading to suppression of the target weed. During the present investigation, cell free culture filtrate (CFCF) of Alternaria alternata was evaluated for its phytotoxicity against a noxious weed Lantana camara. The results of cut shoot, seed- ling and detached leaf bioassays revealed the presence of a toxic metabolite in the CFCF and a significant reduction in chlorophyll and protein content were also noticed. Phytotoxic moiety was further purified and characterized by using solvent partition, thin layer chromatography (TLC), FTIR and 1 H NMR analysis. The acetone extract induced maximum phytotoxic damage at a concentration of 100 µg/ml and TLC purified fraction also exhibited herbicidal potential. The toxic compound was identified as tenuazonic acid upon comparison with FTIR and 1 H NMR spectra. This is the first evidence that confirmed the herbicidal potential of a biorational, tenu- azonic acid was produced by submerged fermentation of A. alternata.

Isolation and characterization of tenuazonic acid produced by alternaria alternata, a potential bioherbicidal agent for control of lantana camara

Chlorophytum borivilianum Sant. et Fernand an endangered herb is valued for several medicinal properties in its tuberous roots. An efficient and reproducible method for inducing in vitro tubers from stem disc explant has been developed. Stem disc possessing shoot buds were induced to develop multiple shoots in Murashige and Skoog (MS) medium supplemented with vitamins, 3% sucrose, 0.8% agar and 5 mg/L benzylaminopurine (BAP). Healthy regenerated shoots were rooted in MS basal medium containing 3% sucrose (w/v), 0.8% agar supplemented with indole-3-acetic acid (2 mg/L). On further sub culturing, the maximum percentage of tuber formation was obtained in growth hormone free half (½) MS liquid media supplemented with vitamins and 1.5% sucrose after 8 to 9 weeks. The saponin contents of the in vitro and in vivo raised tubers were qualitatively and quantitatively analyzed by highperformance liquid chromatography (HPLC) and liquid chromatography electrospray ionization mass spectrometry (LC-ESI-MS). There was a significant similarity in the saponins in both tubers. The in vitro raised tubers showed similar high metabolite content than in vivo grown tubers which is required for medicinal applications. The rooted plantlets were transferred to peat and sand (2:1) with more than 80% success. This is the 1 st report of in vitro tuber formation and secondary metabolites screening of C. borivilianum. This work will give a strong impetus to the pharmaceutical and neutraceutical sectors.
