TL;DR: It is shown that R-loop-associated chromosomal instability can be induced by the loss of DICER1 function, and targeting R-loops with topoisomerase and PARP inhibitors might be an effective treatment strategy for this deadly disease.

TL;DR: The first human SMO inhibitor resistant medulloblastoma PDX models are reported, which can be used for further preclinical experiments to develop the best strategies to overcome the resistance to SMO inhibitors in patients.

TL;DR: Investigation of other pediatric brain tumor types with high levels of R-loops and in vitro drug screens found PARP inhibitors were identified as the most synergistic combination partners for the topoisomerase inhibitor Irinotecan and the active metabolite SN-38, but normal Astrocytes were not sensitive to these combinations.
