Exome sequencing identifies frequent inactivating mutations in BAP1, ARID1A and PBRM1 in intrahepatic cholangiocarcinomas
Yuchen Jiao,Timothy M. Pawlik,Robert A. Anders,Florin M. Selaru,Mirte M. Streppel,Donald J. Lucas,Noushin Niknafs,Violeta Beleva Guthrie,Anirban Maitra,Pedram Argani,G. Johan A. Offerhaus,Juan Carlos Roa,Lewis R. Roberts,Gregory J. Gores,Irinel Popescu,Sorin Alexandrescu,Simona Dima,Matteo Fassan,Michele Simbolo,Andrea Mafficini,Paola Capelli,Rita T. Lawlor,Andrea Ruzzenente,Alfredo Guglielmi,Giampaolo Tortora,Filippo de Braud,Aldo Scarpa,William R. Jarnagin,David S. Klimstra,Rachel Karchin,Victor E. Velculescu,Ralph H. Hruban,Bert Vogelstein,Kenneth W. Kinzler,Nickolas Papadopoulos,Laura D. Wood +35 more
TL;DR: Through exomic sequencing of 32 intrahepatic cholangiocarcinomas, frequent inactivating mutations in multiple chromatin-remodeling genes are discovered (including BAP1, ARID1A and PBRM1), and mutation in one of these genes occurred in almost half of the carcinomas sequenced.
Journal ArticleDOI
Intrahepatic Cholangiocarcinoma: An International Multi-Institutional Analysis of Prognostic Factors and Lymph Node Assessment
Mechteld C. de Jong,Hari Nathan,Georgios C. Sotiropoulos,Andreas Paul,Sorin Alexandrescu,Hugo Marques,Carlo Pulitano,Eduardo Barroso,Bryan M. Clary,Luca Aldrighetti,Cristina R. Ferrone,Andrew X. Zhu,Todd W. Bauer,Dustin M. Walters,T. Clark Gamblin,Kevin Nguyen,Ryan S. Turley,Irinel Popescu,Catherine Hubert,Stephanie Meyer,Richard D. Schulick,Michael A. Choti,Jean-François Gigot,Gilles Mentha,Timothy M. Pawlik +24 more
TL;DR: Although tumor size provides no prognostic information, tumor number, vascular invasion, and LN metastasis were associated with survival and should be strongly considered for ICC.
Journal ArticleDOI
A Nomogram to Predict Long-term Survival After Resection for Intrahepatic Cholangiocarcinoma: An Eastern and Western Experience
Omar Hyder,Hugo Marques,Carlo Pulitano,J. Wallis Marsh,Sorin Alexandrescu,Todd W. Bauer,T. Clark Gamblin,Georgios C. Sotiropoulos,Andreas Paul,Eduardo Barroso,Bryan M. Clary,Luca Aldrighetti,Cristina R. Ferrone,Andrew X. Zhu,Irinel Popescu,Jean-François Gigot,Gilles Mentha,Shen Feng,Timothy M. Pawlik +18 more
TL;DR: An international multicenter study of 514 patients who underwent resection for ICC at 13 major hepatobiliary centers in the United States, Europe, and Asia and a nomogram was developed to predict overall survival after resection, suggesting clinical utility to improve individualized predictions of survival for patients undergoing resection.
Journal ArticleDOI
Recurrence After Operative Management of Intrahepatic Cholangiocarcinoma
Omar Hyder,Ioannis Hatzaras,Georgios C. Sotiropoulos,Andreas Paul,Sorin Alexandrescu,Hugo Marques,Carlo Pulitano,Eduardo Barroso,Bryan M. Clary,Luca Aldrighetti,Cristina R. Ferrone,Andrew X. Zhu,Todd W. Bauer,Dustin M. Walters,Ryan T. Groeschl,T. Clark Gamblin,J. Wallis Marsh,Kevin Nguyen,Ryan S. Turley,Irinel Popescu,Catherine Hubert,Stephanie Meyer,Michael A. Choti,Jean-François Gigot,Gilles Mentha,Timothy M. Pawlik +25 more
TL;DR: Recurrence after operative intervention for ICC was common and disease recurred both at intra- and extrahepatic sites with roughly the same frequency, factors such as lymph node metastasis, tumor size, and vascular invasion predict highest risk of recurrence.
Journal ArticleDOI
Management and Outcomes of Patients with Recurrent Intrahepatic Cholangiocarcinoma Following Previous Curative-Intent Surgical Resection
Gaya Spolverato,Yuhree Kim,Sorin Alexandrescu,Hugo Marques,Jorge Lamelas,Luca Aldrighetti,T. Clark Gamblin,Shishir K. Maithel,Carlo Pulitano,Todd W. Bauer,Feng Shen,George A. Poultsides,Thuy B. Tran,J. Wallis Marsh,Timothy M. Pawlik,Timothy M. Pawlik +15 more
TL;DR: The treatment of patients with recurrent intrahepatic cholangiocarcinoma following curative-intent surgery was characterized and therapy-specific outcomes defined, and recurrence following resection of ICC was common.
