Journal ArticleDOI
Repressive and active histone methylation mark distinct promoters in human and mouse spermatozoa
Urszula Brykczynska,Mizue Hisano,Serap Erkek,Liliana Ramos,Edward J Oakeley,Edward J Oakeley,Tim Roloff,Christian Beisel,Dirk Schübeler,Michael B. Stadler,Antoine H.F.M. Peters +10 more
TL;DR: Two modifications important for Trithorax- and Polycomb-mediated gene regulation have methylation-specific distributions at regulatory regions in human spermatozoa, compatible with a role for Polycomb in repressing somatic determinants across generations, potentially in a variegating manner.
Journal ArticleDOI
PRC1 and Suv39h specify parental asymmetry at constitutive heterochromatin in early mouse embryos
Mareike Puschendorf,Rémi Terranova,Erwin Boutsma,Xiaohong Mao,Xiaohong Mao,Kyoichi Isono,Urszula Brykczynska,Carolin Kolb,Arie P. Otte,Haruhiko Koseki,Stuart H. Orkin,Maarten van Lohuizen,Antoine H.F.M. Peters +12 more
TL;DR: It is concluded that in early embryos, Suv39h-mediated H3K9me3 constitutes the dominant maternal transgenerational signal for pericentric heterochromatin formation and in absence of this signal, PRC1 functions as the default repressive back-up mechanism.
Journal ArticleDOI
Impaired function of primitive hematopoietic cells in mice lacking the Mixed-Lineage-Leukemia homolog MLL5.
Vikas Madan,Babita Madan,Urszula Brykczynska,Frederic Zilbermann,Kevin Hogeveen,Konstanze Döhner,Hartmut Döhner,Odile Weber,Carmen Blum,Hans Reimer Rodewald,Paolo Sassone-Corsi,Antoine H.F.M. Peters,Hans Jörg Fehling +12 more
TL;DR: In vivo findings establish several nonredundant functions for Mll5, including an essential role in regulating proliferation and functional integrity of hematopoietic stem/progenitor cells, and suggest a role in epigenetic gene regulation.
Journal ArticleDOI
Cbx2 Targets PRC1 to Constitutive Heterochromatin in Mouse Zygotes in a Parent-of-Origin-Dependent Manner
Mathieu Tardat,Mareike Albert,Rico Kunzmann,Rico Kunzmann,Zichuan Liu,Lilia Kaustov,Raphael Thierry,Shili Duan,Urszula Brykczynska,Urszula Brykczynska,Cheryl H. Arrowsmith,Antoine H.F.M. Peters,Antoine H.F.M. Peters +12 more
TL;DR: A PRC1 targeting mechanism that relies on Cbx2 and Hp1β is identified that separates by a short linker function together to integrate H 3K9me3/HP1 and H3K27me3 states.
Journal ArticleDOI
Genetic Vasectomy—Overexpression of Prm1-EGFP Fusion Protein in Elongating Spermatids Causes Dominant Male Sterility in Mice
Sabine Haueter,Miyuri Kawasumi,Igor N. Asner,Urszula Brykczynska,Paolo Cinelli,Stefan Moisyadi,Kurt Bürki,Antoine H.F.M. Peters,Pawel Pelczar +8 more
TL;DR: The “genetic vasectomy system” was improved by genetically linking the dominant male sterility to ubiquitous EGFP expression in the soma as an easy phenotypic marker enabling rapid genotyping of transgenic males and females.
