T Lymphocyte-Directed Gene Therapy for ADA− SCID: Initial Trial Results After 4 Years
R. Michael Blaese,Kenneth W. Culver,A. Dusty Miller,Charles S. Carter,Thomas A. Fleisher,Mario Clerici,Gene M. Shearer,Lauren A. Chang,Yawen Chiang,Paul Tolstoshev,Jay J. Greenblatt,Steven A. Rosenberg,Harvey G. Klein,Melvin Berger,Craig A. Mullen,W. Jay Ramsey,Linda M. Muul,Richard A. Morgan,W. French Anderson +18 more
TL;DR: In this article, a clinical trial was started using retroviral-mediated transfer of the ADA gene into the T cells of two children with severe combined immunodeficiency (ADA-SCID).
Journal ArticleDOI
Gene Transfer into Humans — Immunotherapy of Patients with Advanced Melanoma, Using Tumor-Infiltrating Lymphocytes Modified by Retroviral Gene Transduction
Steven A. Rosenberg,Paul Aebersold,Kenneth Cornetta,Attan Kasid,Richard A. Morgan,Robert C. Moen,Karson E,Michael T. Lotze,James Chih-Hsin Yang,Suzanne L. Topalian,Maria J. Merino,Kenneth W. Culver,A. Dusty Miller,R. Michael Blaese,W. French Anderson +14 more
TL;DR: These studies demonstrate the feasibility and safety of using retroviral gene transduction for human gene therapy and have implications for the design of TIL with improved antitumor potency, as well as for the possible use of lymphocytes for the gene therapy of other diseases.
Journal ArticleDOI
A nanoliter-scale nucleic acid processor with parallel architecture.
Jong Wook Hong,Vincent Studer,Vincent Studer,Giao B. Hang,W. French Anderson,W. French Anderson,Stephen R. Quake +6 more
TL;DR: These microfluidic chips for automated nucleic acid purification from small numbers of bacterial or mammalian cells are developed, illustrating how highly parallel microfluidity architectures can be constructed to perform integrated batch-processing functionalities for biological and medical applications.
Journal ArticleDOI
Genetic marking shows that Ph+ cells present in autologous transplants of chronic myelogenous leukemia (CML) contribute to relapse after autologous bone marrow in CML.
Albert B. Deisseroth,Z. Zu,David F. Claxton,E. G. Hanania,Siqing Fu,Debra Ellerson,Leah Goldberg,Michael Thomas,Karen Janicek,W. French Anderson,Jeane P. Hester,Martin Korbling,April G. Durett,Robert C. Moen,Ronald J. Berenson,Shelly Heimfeld,Joy Hamer,Leslie Calvert,Patricia Tibbits,Moshe Talpaz,Hagop M. Kantarjian,Richard E. Champlin,Christopher L. Reading +22 more
TL;DR: It is shown that it is possible to use a replication-incompetent safety-modified retrovirus in order to introduce DNA sequences into the hematopoietic cells of patients undergoing autologous bone marrow transplantation.
Journal ArticleDOI
Improved methods of retroviral vector transduction and production for gene therapy.
Hitoshi Kotani,Perry Newton,Shuyuan Zhang,Yawen L. Chiang,Edward Otto,L. H. Weaver,R. Michael Blaese,W. French Anderson,Gerard J. McGarrity +8 more
TL;DR: In studies on transduction by retroviral vectors, centrifugation of vector supernatants onto target cells significantly increased transduction efficiency as measured by vector titration for G418 resistance, fluorescence-activated cell sorting (FACS), and polymerase chain reaction (PCR) analyses.
