Journal ArticleDOI

Microenvironment-induced PTEN loss by exosomal microRNA primes brain metastasis outgrowth

TL;DR: A remarkable plasticity of PTEN expression in metastatic tumour cells in response to different organ microenvironments is demonstrated, underpinning an essential role of co-evolution between the metastatic cells and their microenvironment during the adaptive metastatic outgrowth.
Journal ArticleDOI

Combating trastuzumab resistance by targeting SRC, a common node downstream of multiple resistance pathways

TL;DR: It is found that SRC is activated in both acquired and de novo trastuzumab-resistant cells and a novel mechanism of SRC regulation involving dephosphorylation by PTEN is uncovered, suggesting the potential clinical application of this strategy to overcome trastzumab resistance.
Journal ArticleDOI

Src Family Kinases as Novel Therapeutic Targets to Treat Breast Cancer Brain Metastases

TL;DR: A critical role for Src activation in promoting brain metastasis in a preclinical model of breast cancer is revealed and how Src-targeting combinatorial regimens can treat HER2(+) brain metastases in this model is shown.
Journal ArticleDOI

Mitotic Deregulation by Survivin in ErbB2-Overexpressing Breast Cancer Cells Contributes to Taxol Resistance

TL;DR: It is found that ErbB2-overexpressing cells expressed higher levels of survivin in multiple breast cancer cell lines and patient samples, which correlated with the increased resistance to Taxol-induced apoptosis and combination treatment of Taxol with PI3K/Akt and Src inhibitor led to increased apoptosis.
