Journal ArticleDOI
Angiotensin-converting enzyme 2 is an essential regulator of heart function
Michael A. Crackower,Renu Sarao,Renu Sarao,Gavin Y. Oudit,Gavin Y. Oudit,Chana Yagil,Ivona Kozieradzki,Ivona Kozieradzki,Sam E. Scanga,Antonio J. Oliveira-dos-Santos,Joan da Costa,Liyong Zhang,York Pei,James W. Scholey,Carlos M. Ferrario,Armen S. Manoukian,Mark C. Chappell,Peter H. Backx,Peter H. Backx,Yoram Yagil,Josef M. Penninger +20 more
TL;DR: These genetic data for ACE2 show that it is an essential regulator of heart function in vivo and targeted disruption of ACE2 in mice results in a severe cardiac contractility defect, increased angiotensin II levels, and upregulation of hypoxia-induced genes in the heart.
Journal ArticleDOI
Anti-TGF-β1 Antibody Therapy in Patients with Diabetic Nephropathy.
James Voelker,Paul Berg,Matthew J. Sheetz,Kevin L. Duffin,Tong Shen,Brian A. Moser,Tom Greene,Samuel S. Blumenthal,Ivan Rychlik,Yoram Yagil,Philippe Zaoui,Julia B. Lewis +11 more
TL;DR: TGF-β1 mAb added to renin-angiotensin system inhibitors did not slow progression of diabetic nephropathy and the trial was terminated 4 months early for futility on the basis of their recommendation.
Journal ArticleDOI
ACE2 expression and activity are enhanced during pregnancy
TL;DR: During pregnancy, the placentas, in particular, but also the uterus, constitute important sources of ACE2, in addition to its normal production in the kidney, leading to an estimated twofold increase in total ACE2 activity.
Journal ArticleDOI
Pyridorin in Type 2 Diabetic Nephropathy
Edmund J. Lewis,Tom Greene,Samuel Spitalewiz,Samuel S. Blumenthal,Tomas Berl,Lawrence G. Hunsicker,Marc A. Pohl,Richard D. Rohde,Itamar Raz,Yair Yerushalmy,Yoram Yagil,Tommy Herskovits,Robert C. Atkins,Anne T. Reutens,David K. Packham,Julia B. Lewis +15 more
TL;DR: This trial failed to detect an effect of Pyridorin on the progression of serum creatinine at 1 year, although it suggests that patients with less renal impairment might benefit, and there was no evidence of a significant treatment effect in the middle or upper tertiles.
Journal ArticleDOI
Hypothesis. ACE2 Modulates Blood Pressure in the Mammalian Organism
Yoram Yagil,Chana Yagil +1 more
TL;DR: It is now well established that hyperactivation of the RAS invariably leads to hypertension and to a number of other adverse cardiovascular effects that can be, at least in part, prevented by ACE inhibition or angiotensin receptor blockade.
