Journal ArticleDOI
Identification of an antimalarial synthetic trioxolane drug development candidate.
Jonathan L. Vennerstrom,Sarah Arbe-Barnes,Reto Brun,Susan A. Charman,Francis C. K. Chiu,Jacques Chollet,Yuxiang Dong,Arnulf Dorn,Daniel Hunziker,Hugues Matile,Kylie Anne McIntosh,Maniyan Padmanilayam,Josefina Santo Tomas,Christian Scheurer,Bernard Scorneaux,Yuanqing Tang,Heinrich Urwyler,Sergio Wittlin,William N. Charman +18 more
TL;DR: Here it is described how a synthetic peroxide antimalarial drug development candidate was identified in a collaborative drug discovery project.
Journal ArticleDOI
Spiro and dispiro-1,2,4-trioxolanes as antimalarial peroxides: charting a workable structure-activity relationship using simple prototypes.
Yuxiang Dong,Jacques Chollet,Hugues Matile,Susan A. Charman,Francis C. K. Chiu,William N. Charman,Bernard Scorneaux,Heinrich Urwyler,Josefina Santo Tomas,Christian Scheurer,Christopher Snyder,Arnulf Dorn,Xiaofang Wang,Jean M. Karle,Yuanqing Tang,Sergio Wittlin,Reto Brun,Jonathan L. Vennerstrom +17 more
TL;DR: The trioxolanes were substantially less effective in a standard oral suspension formulation compared to a solubilizing formulation and were more active when administered subcutaneously than orally, both of which suggest substantial biopharmaceutical liabilities.
Journal ArticleDOI
Dispiro-1,2,4-trioxane Analogues of a Prototype Dispiro-1,2,4-trioxolane: Mechanistic Comparators for Artemisinin in the Context of Reaction Pathways with Iron(II)
Yuanqing Tang,Yuxiang Dong,Xiaofang Wang,Kamaraj Sriraghavan,James K. Wood,Jonathan L. Vennerstrom +5 more
TL;DR: It is suggested that formation of either primary or secondary carbon-centered radicals is a necessary but insufficient criterion for antimalarial activity of 1 and synthetic peroxides.
Journal ArticleDOI
The structure-activity relationship of the antimalarial ozonide arterolane (OZ277).
Yuxiang Dong,Sergio Wittlin,Kamaraj Sriraghavan,Jacques Chollet,Susan A. Charman,William N. Charman,Christian Scheurer,Heinrich Urwyler,Josefina Santo Tomas,Christopher Snyder,Darren J. Creek,Julia Morizzi,Maria Koltun,Hugues Matile,Xiaofang Wang,Maniyan Padmanilayam,Yuanqing Tang,Arnulf Dorn,Reto Brun,Jonathan L. Vennerstrom +19 more
TL;DR: Five new ozonides with antimalarial efficacy and ADME profiles superior or equal to that of arterolane were identified.
Journal ArticleDOI
Synthesis of tetrasubstituted ozonides by the Griesbaum coozonolysis reaction: diastereoselectivity and functional group transformations by post-ozonolysis reactions.
TL;DR: It is evident that these tetrasubstituted ozonide isomers are quite stable to triphenylphosphine, borohydrides, hydrazine, alkyllithiums, Grignard reagents, mercaptides, and aqueous KOH as illustrated by the synthesis of amine, alcohol, acid, ester, ether, sulfide, sulfone, and heterocycle-functionalized ozonides by a wide range of
